目的:研究肉桂酸(CINN)对大鼠骨髓间充质干细胞(BMSCs)增殖和向成骨细胞分化的影响。方法实验分为CINN组和对照组。 CINN组采用CINN处理BMSCs,对照组细胞培养基不加任何药物处理。显微镜观察细胞形态变化,四甲基偶氮唑蓝法( MTT)测定细胞增殖能力,流式细胞仪分析细胞周期,茜素红染色观察BMSCs向成骨细胞的分化能力, RT-PCR及放免方法检测骨钙素( BGP)的表达。结果与对照组比较,CINN组BMSCs增殖能力显著下降,细胞周期出现向G0/G1期移行的特征性动力学改变。 CINN作用14 d后钙化结节形成,且骨钙素的表达明显升高。结论肉桂酸可抑制大鼠BMSCs增殖,促进其向成骨细胞分化。

Object ive It is to investigate the effects of cinnamic acid ( CINN) on the proliferation and differentiation of bone mesenchymal stem cells (BMSCs) in rats.Methods The experiment was divided into CINN group and control group . BMSCs were treated with CINN in CINN group , and the cells were treated with no drugs in control group .The morphological changes of cells were observed by microscope .The rate of growth suppression was evaluated by MTT assay .Calcified nodules were observed using alizarin red staining .Cell cycle was analyzed by flow cytometry .The expression of BGP was examined with RT-PCR assay and radio immunoassay .Results The cell proliferating ability decreased significantly in CINN group com-pared with the control group .Flow cytometry analysis showed that CINN increased the cell numbers in G 0/G1 phase signifi-cantly(P<0.01).The formation of calcified nodules and the expression of BGP increased after treatment with CINN for 14 days.Conclusion CINN can inhibit the proliferation of BMSCs and induce the differentiation of BMSCs into osteoblasts .
