探讨含脑血康颗粒的培养基对脑出血患者外周血中提取的血管内皮祖细胞(EPCs)的数量及功能的影响。方法采用密度梯度离心法分离出脑出血患者外周血的EPCs,加入含脑血康颗粒的相应培养基中,并根据培养基中脑血康颗粒浓度0,10,50,100 mg/mL分为对照组、低剂量组、中剂量组和高剂量组,分别检测培养基孵育7 d后EPCs的数量及细胞功能。结果与正常组相比,脑出血组的外周血EPCs数目及黏附、迁移、增殖及体外血管形成能力均明显降低(P均<0.05);与对照组相比,脑血康各组外周血的EPCs增殖能力及黏附、迁移及体外血管形成能力均明显提高(P均<0.05)。结论脑血康颗粒能明显增加脑出血患者外周血EPCs的数量,并且改善其功能,增强EPCs的增殖、分化、黏附和迁移能力,能够更快形成血管样结构。

Objective:It is to explore the effects of culture medium containing liquid medicine of Naoxuekang granules on the number and the function of endothelial progenitor cells ( EPCs ) isolated from peripheral blood in patients with cerebral hemorrhage .M ethods EPCs was isolated from peripheral blood of patients with cerebral hemorrhage by the method of density gradient centrifugation .According to the Naoxuekang granules concentration (0,10,50,100 mg/mL) added to culture medi-um, the cells were divided into control group , low dose group , middle dose group and high dose group respectively .Those groups were incubated for 7 days,then the number of EPCs and their impact on cell function was observed .Results In cerebral hemorrhage group compared with normal group ,it is observed that the number of EPCs in peripheral blood and the ability of ad-hesion, migration, proliferation and angiogenesis in vitro decreased obviously (P<0.05).In the Naoxuekang groups compared with control group , the number of EPCs in peripheral blood and the ability of adhesion , migration , proliferation and angiogene-sis in vitro increased significantly ( P<0.05).Conclusion Naoxuekang granules can significantly increase the number of EPCs in patients with cerebral hemorrhage , and improve their capabilities of proliferation , differentiation , adhesion , migration and angiogenesis , thus to formulate angiogenesis more faster .
