目的:研究血管内皮生长因子( vascular endothelial growth factor,VEGF)及炎症相关细胞因子在实验性自身免疫性脑脊髓炎( experimental autoimmune encephalomyelitis,EAE)大鼠血清中的表达及探讨与疾病进展的关系。方法:雌性Wistar大鼠30只随机分为模型组20只及对照组10只,由豚鼠脊髓匀浆及百日咳菌液作为抗原免疫模型组大鼠制作EAE模型,于发病高峰时取得大鼠血清及脑脊髓标本,用酶联免疫吸附法( enzyme linked immunosorbent assay,ELISA)测血清VEGF、白细胞介素-4(IL-4)、白细胞介素-10(IL-10)、干扰素-γ(IFN-γ)及髓鞘碱性蛋白(MBP)水平,组织学切片观察脑及脊髓病理变化。结果:(1)模型组大鼠全部发病,可见明显炎细胞浸润及髓鞘脱失改变;(2)VEGF在模型组大鼠血清中表达较对照组有明显增高(P<0.05),且VEGF水平的升高与动物模型发病临床评分呈显著正相关,与血清IFN-γ、MBP水平升高及IL-4水平降低亦相一致。结论: EAE血清VEGF水平与临床症状加重、促炎因子增高、抑炎因子降低及髓鞘破坏程度一致,提示VEGF可能通过与炎症因子协同作用参与了EAE的病变进展。

목적:연구혈관내피생장인자( vascular endothelial growth factor,VEGF)급염증상관세포인자재실험성자신면역성뇌척수염( experimental autoimmune encephalomyelitis,EAE)대서혈청중적표체급탐토여질병진전적관계。방법:자성Wistar대서30지수궤분위모형조20지급대조조10지,유돈서척수균장급백일해균액작위항원면역모형조대서제작EAE모형,우발병고봉시취득대서혈청급뇌척수표본,용매련면역흡부법( enzyme linked immunosorbent assay,ELISA)측혈청VEGF、백세포개소-4(IL-4)、백세포개소-10(IL-10)、간우소-γ(IFN-γ)급수초감성단백(MBP)수평,조직학절편관찰뇌급척수병리변화。결과:(1)모형조대서전부발병,가견명현염세포침윤급수초탈실개변;(2)VEGF재모형조대서혈청중표체교대조조유명현증고(P<0.05),차VEGF수평적승고여동물모형발병림상평분정현저정상관,여혈청IFN-γ、MBP수평승고급IL-4수평강저역상일치。결론: EAE혈청VEGF수평여림상증상가중、촉염인자증고、억염인자강저급수초파배정도일치,제시VEGF가능통과여염증인자협동작용삼여료EAE적병변진전。
Objective:To study the expression of VEGF and inflammation related cytokines in the serum of EAE rats and investigate the relationship with the progression of EAE. Methods:30 female Wistar rats were randomly divided into the model group(20)and the control group(10). Model group had been immunized with guinea pig spinal cord homogenate and pertussis bacilli. Serum,brain and spinal cord specimens were obtained at peak of the disease. VEGF,IL-4,IL-10,IFN-γ and MBP levels in serum were measured by ELISA. Pathological changes in the brain and spinal cord were evaluated by histological sections. Results:(1)All model rats had symp-toms. Inflammatory cells infiltration and demyelination change were obvious in model rats. (2)The expression of VEGF in the serum of model group was significantly higher than the control group(P<0. 05). The increasing of VEGF was significantly correlated with clini-cal scores,and also consistent with the increasing of IFN-γ,MBP,the reducing of IL-4. Conclusion:VEGF levels in EAE serum were consistent with its clinical symptoms,the increasing of pro-inflammatory cytokines,the reducing of inhibition cytokines and the damage of myelin. VEGF may be synergistic with the inflammatory cytokines involved in the disease progression of EAE.
