目的:比较细胞因子诱导的杀伤细胞( CIK)及树突状细胞-CIK( DC-CIK)在体外培养特性及收获后室温放置不同时间的存活情况。方法选择中晚期恶性肿瘤住院患者50例,分为CIK组和DC-CIK组,采血前检测患者的血常规,采血后记录分选淋巴细胞的数量和收获淋巴细胞总数,在培养的第15天进行流式细胞术检测CIK及DC-CIK细胞的表面标志CD3、CD4、CD8、CD56的表达,收获后胎盘蓝染色法检测细胞在室温下放置不同时间存活率的变化。结果两组初始淋巴细胞数、收获淋巴细胞总数相比差异无显著性,扩增倍数DC-CIK组显著高于CIK组;CD3+、CD8+、CD3+CD8+和CD3+CD56+T细胞比例DC-CIK组显著高于CIK组,两组细胞活率随时间延长而降低,2 h细胞活率与初始有显著性差异,4 h细胞活率低于95%。相同时间两组间细胞活率比较差异无显著性。结论 DC-CIK细胞比CIK细胞增殖更快,效应细胞比例更高,因而具有更高的生物活性。收获后细胞在室温下放置,随时间延长活率逐渐下降,所以细胞收获后应尽快回输给患者。

목적:비교세포인자유도적살상세포( CIK)급수돌상세포-CIK( DC-CIK)재체외배양특성급수획후실온방치불동시간적존활정황。방법선택중만기악성종류주원환자50례,분위CIK조화DC-CIK조,채혈전검측환자적혈상규,채혈후기록분선림파세포적수량화수획림파세포총수,재배양적제15천진행류식세포술검측CIK급DC-CIK세포적표면표지CD3、CD4、CD8、CD56적표체,수획후태반람염색법검측세포재실온하방치불동시간존활솔적변화。결과량조초시림파세포수、수획림파세포총수상비차이무현저성,확증배수DC-CIK조현저고우CIK조;CD3+、CD8+、CD3+CD8+화CD3+CD56+T세포비례DC-CIK조현저고우CIK조,량조세포활솔수시간연장이강저,2 h세포활솔여초시유현저성차이,4 h세포활솔저우95%。상동시간량조간세포활솔비교차이무현저성。결론 DC-CIK세포비CIK세포증식경쾌,효응세포비례경고,인이구유경고적생물활성。수획후세포재실온하방치,수시간연장활솔축점하강,소이세포수획후응진쾌회수급환자。
Objective To observe and compare characteristics and survival rate after harvest of CIK and DC-CIK cells. Methods A total of 50 patients with advanced tumor were enrolled in this study and they were divided into two groups( CIK or DC-CIK cells). Routine blood cells counts were analyzed before the culture of CIK or DC-CIK cells,and lymphocytes were counted after collection of blood samples and har-vest,the subtypes of T cells were analyzed by flow cytometry,and Taipan blue coloring had been used for testing survival rate. Results The difference in total number of initial lymphocytes and harvested CIK or DC-CIK cells was not significant,but the amplification of DC-CIK group was higher then that of CIK group. The percentage of CD3 +,CD8 +,CD3 +CD8 + and CD3 +CD56 + T cells in DC-CIK group was significantly higher than that of CIK group. The cell survival rate was decreasing with the time went on outside the incubator,there was significant difference between 2 h and initial survival rate. The survival rate of cells was less than 95% at the end of 4 h. There was no significant difference in survival rate between these two groups at the same time. Conclusion DC-CIK cells have higher killing activity and they can grow more rapidly than CIK cells and have higher ratio of effective cells. After harvest,the survival rate of both CIK cells and DC-CIK cells is decreased as the times going on outside the incubator. Both CIK cells or DC-CIK cells should transfuse to patients as quickly as possible.
