目的:初步探讨正常软骨与骨性关节炎(OA)软骨中炎性细胞因子---白介素(IL)-6、缺氧诱导因子(HIF)-α、基质金属蛋白酶(MMP)-3等的表达差异。方法收集2013年1~6月该院收治的7例OA患者(观察组)的软骨标本及9例其他患者(对照组)的健康软骨标本。采用苏木精-伊红(HE)染色法观察软骨病理学表现,采用蛋白质印迹法检测两组患者软骨中IL-6、HIF-α、MMP-3的表达情况。结果观察组患者软骨中软骨细胞减少,细胞簇集,可见软骨细胞变性、坏死,软骨厚度变薄,基质染色不均匀,有异染性;IL-6、HIF-α、MMP-3的表达明显高于对照组,差异均有统计学意义(P<0.05)。结论 IL-6、HIF-α、MMP-3可能在OA的发病中具有重要作用,有望为临床治疗OA开辟一条重要途径。

목적:초보탐토정상연골여골성관절염(OA)연골중염성세포인자---백개소(IL)-6、결양유도인자(HIF)-α、기질금속단백매(MMP)-3등적표체차이。방법수집2013년1~6월해원수치적7례OA환자(관찰조)적연골표본급9례기타환자(대조조)적건강연골표본。채용소목정-이홍(HE)염색법관찰연골병이학표현,채용단백질인적법검측량조환자연골중IL-6、HIF-α、MMP-3적표체정황。결과관찰조환자연골중연골세포감소,세포족집,가견연골세포변성、배사,연골후도변박,기질염색불균균,유이염성;IL-6、HIF-α、MMP-3적표체명현고우대조조,차이균유통계학의의(P<0.05)。결론 IL-6、HIF-α、MMP-3가능재OA적발병중구유중요작용,유망위림상치료OA개벽일조중요도경。
Objective To prelininarily investigate the expression levels of interleukin(IL)-6,hypoxia-inducible factor (HIF)-αand matrix metalloproteinases(MMP)-3 in inflammatory factor of osteoarthritis(OA) cartilages and normal cartilages. Meth-ods Specimens of human articular cartilage were collected from 7 OA patients(observation group) and 9 other patients(control group). HE staining was adopted to observe the histologic structure of cartilage,and expressions of IL-6,HIF-α,MMP-3 were detected by Western blotting. Results The number of chondrocytes in the observation group delined and clustered,showing chondrocytes degeneration,and necrosis,thin of cartilage,uneven of matrix staining and metachromia,and the expressions of IL-6,HIF-αand MMP-3 were obviously higher than those in the control group,and the differences all had statistical significance (P<0.05). Con-clusion IL-6,HIF-α,MMP-3 may play an important role in OA,which are expected to open up a new way to treat OA in clinic.
