目的:研究γ氨基丁酸(GABA)对野百合碱(MCT )诱导肺动脉高压大鼠的治疗作用,并检测血管内皮生长因子(VEGF)、基质金属蛋白酶-9(MMP-9)表达水平的变化。方法应用随机数字表法将30只雄性SD大鼠随机分3组,分别为正常对照组(对照组)、MCT模型组(模型组)、γ氨基丁酸干预组(干预组),每组10只。观测平均右心室压(mRVP),计算右心室肥厚指数(RVHI),测定肺小动脉管壁厚度与动脉外直径的百分比(WT%)及管壁面积与血管总面积的百分比(WA%),免疫组织化学染色法观察MMP-9、实时荧光定量PCR(FQ-PCR)检测VEGF mRNA在肺组织中的表达。结果干预组mRVP、RVHI、WT%、WA%、MMP-9蛋白和VEGF mRNA的表达水平明显高于对照组(P<0.05);而低于模型组(P<0.05)。结论 GABA可抑制野百合碱诱导的肺动脉高压模型大鼠肺组织VEGF mRNA和MMP-9蛋白的表达,对肺动脉高压有治疗作用。
目的:研究γ氨基丁痠(GABA)對野百閤堿(MCT )誘導肺動脈高壓大鼠的治療作用,併檢測血管內皮生長因子(VEGF)、基質金屬蛋白酶-9(MMP-9)錶達水平的變化。方法應用隨機數字錶法將30隻雄性SD大鼠隨機分3組,分彆為正常對照組(對照組)、MCT模型組(模型組)、γ氨基丁痠榦預組(榦預組),每組10隻。觀測平均右心室壓(mRVP),計算右心室肥厚指數(RVHI),測定肺小動脈管壁厚度與動脈外直徑的百分比(WT%)及管壁麵積與血管總麵積的百分比(WA%),免疫組織化學染色法觀察MMP-9、實時熒光定量PCR(FQ-PCR)檢測VEGF mRNA在肺組織中的錶達。結果榦預組mRVP、RVHI、WT%、WA%、MMP-9蛋白和VEGF mRNA的錶達水平明顯高于對照組(P<0.05);而低于模型組(P<0.05)。結論 GABA可抑製野百閤堿誘導的肺動脈高壓模型大鼠肺組織VEGF mRNA和MMP-9蛋白的錶達,對肺動脈高壓有治療作用。
목적:연구γ안기정산(GABA)대야백합감(MCT )유도폐동맥고압대서적치료작용,병검측혈관내피생장인자(VEGF)、기질금속단백매-9(MMP-9)표체수평적변화。방법응용수궤수자표법장30지웅성SD대서수궤분3조,분별위정상대조조(대조조)、MCT모형조(모형조)、γ안기정산간예조(간예조),매조10지。관측평균우심실압(mRVP),계산우심실비후지수(RVHI),측정폐소동맥관벽후도여동맥외직경적백분비(WT%)급관벽면적여혈관총면적적백분비(WA%),면역조직화학염색법관찰MMP-9、실시형광정량PCR(FQ-PCR)검측VEGF mRNA재폐조직중적표체。결과간예조mRVP、RVHI、WT%、WA%、MMP-9단백화VEGF mRNA적표체수평명현고우대조조(P<0.05);이저우모형조(P<0.05)。결론 GABA가억제야백합감유도적폐동맥고압모형대서폐조직VEGF mRNA화MMP-9단백적표체,대폐동맥고압유치료작용。
Objective To study the therapeutical effect of γ-aminobutyric acid on monocrotaline(MCT )induced pulmonary hy-pertension rats ,and to elucidate the expression of VEGF and MMP-9 .Methods Thirty SD rats were randomly divided into 3 groups :a normal control group(control group) ,a MCT-induced pulmonary hypertension group(model control group) ,and anγ-ami-nobutyric acid treatment group(treatment group) .The mean right ventricular pressure(mRVP)were detected ,the right ventricular hypertrophy index(RVHI)were measured ,WT% and WA% were evaluated ,and the expression of VEGF mRNA in the lung tissue and MMP-9 were detected wtih FQ-PCR and immunohistochemical staining method respectively .Results mRVP ,RVHl ,WT% , WA% and the expression of MMP-9 and VEGF mRNA of treatment group were lower than those in the model control group(P<0 .05) ,but higher than the control group(P<0 .05) .Conclusion GABA has a therapeutic effect on pulmonary hypertension rats through regulating the expression of VEGF mRNA and MMP-9 protein .
