[目的]研究缺氧护理干预对人宫颈癌 HeLa 细胞周期和存活等的影响,以期为临床肿瘤护理新措施的制定提供依据。[方法] HeLa 细胞按常规用DMEM高糖培养基培养;制备1000μM的二氯化钴储存液后,分别构建50μM、100μM及200μM的二氯化钴浓度模拟轻度缺氧、中度缺氧及重度缺氧的环境,37℃孵育24 h后按流式细胞检测程序进行细胞周期和存活率测定及克隆形成能力测定。[结果]重度缺氧能引起 HeLa 细胞处于 G1期的细胞比例明显下降,处于 G2/M 期的细胞比例明显升高;重度缺氧还能造成 H eLa 细胞存活率降低;缺氧的严重程度与 H eLa 细胞克隆形成能力的抑制程度呈正相关。[结论]缺氧能严重影响 H eLa 细胞的生长和存活,护理干预可据此进行针对性的新措施制定和临床研究,以便进行更有效的宫颈癌病人个性化护理。

Obj ective:To study the influence of hypoxia nursing interven-tion on cycle and survival of HeLa cells in human cervical carcinoma,in order to provide the basis for formulating new measures of clinical oncol-ogy nursing.Methods:The HeLa cells were cultured in conventional DMEM high glucose medium;after 1 000μM Two cobalt chloride storage liquid were prepared,it respectively constructed two cobalt chloride con-centration of 50μM,100μM and 200μM to simulate mild hypoxia,mod-erate hypoxia and severe hypoxia environment.After incubating for 24 h at 3 7 ℃,then the cell cycle,cell survival rate and clone forming ability were measured according to the flow cytometry detection program.Re-sults:Severe hypoxia can obviously induce the percentage of G1 phase cells in HeLa cells decent,but the proportion of cells in G2/M phase in-creased significantly;severe hypoxia can lead to the survival rate of HeLa cell decent;the severity of hypoxia was positively correlated with the de-gree of inhibition of HeLa cell clone forming ability.Conclusion:Hypoxia can seriously affect the growth and survival of HeLa cells.According to this,the targeted new measures development and clinical research could be carried out in nursing intervention,in order to carry out more effec-tively personalized nursing for patients with cervical cancer.
