目的:对 HBV-DNA 自配室内质控品的配置进行改进、优化,使 HBV-DNA 自配室内质控品起到稳定作用。方法采用实时荧光定量 PCR 技术对收集的阳性混合血清和阴性混合血清分别进行灭活和检测,配制2个水平的室内质控血清,观察其降解程度。结果自配血清质控品过程中采取血清灭活措施后质控品的稳定性提高,同时失控率较优化前大幅下降,自配质控品血清进行一年的累积以后仍能够得到稳定的均值、s 及 CV,而不需做相关参数的调整。结论此种方法稳定性好,操作简便,节省成本,便于推广。

목적:대 HBV-DNA 자배실내질공품적배치진행개진、우화,사 HBV-DNA 자배실내질공품기도은정작용。방법채용실시형광정량 PCR 기술대수집적양성혼합혈청화음성혼합혈청분별진행멸활화검측,배제2개수평적실내질공혈청,관찰기강해정도。결과자배혈청질공품과정중채취혈청멸활조시후질공품적은정성제고,동시실공솔교우화전대폭하강,자배질공품혈청진행일년적루적이후잉능구득도은정적균치、s 급 CV,이불수주상관삼수적조정。결론차충방법은정성호,조작간편,절성성본,편우추엄。
Objective To improve,optimize the method of controlling HBV-DNA serum stability indoor as to increase the stability of HBV-DNA self-made internal quality control products.Methods Real-time PCR technology were used to inactivate and test the mixed collection of positive pooled serum and negative serum,two levels of inter-nal quality control serum was prepared to observe the degree of degradation.Results Self-made quality control serum product was improved stability after inactivated.it′s could be keep one year and still get a stable average,deviation and Standard deviation.Conclusion This method could be a simple,cost-saving,good stability methods.
