目的:观察齐墩果酸(OA)预处理对肝脏缺血再灌注损伤中细胞凋亡的影响,探讨齐墩果酸预处理对肝脏保护作用的机制。方法雄性 SD 大鼠40只,随机分为假手术组(SH 组)、缺血再灌注组(IR 组)、0.5%羧甲基纤维素钠组(CM 组)和齐墩果酸预处理组(齐墩果酸组),齐墩果酸组以100 mg/kg 的齐墩果酸混悬液,SH 和 IR 组以相同容积的水,CM 组以相同容积的0.5%CMC-Na 分别每日灌胃1次,连续7 d。第8天建立70%肝脏缺血再灌注模型,肝脏缺血60 min 后行再灌注3 h。检测大鼠血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)和LDH 水平;检测肝组织内丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽(GSH)水平变化;通过 HE 染色观察肝组织形态,利用 Tunel 染色检测肝组织凋亡状态;Western blot 检测肝脏组织 caspase3和 p53蛋白表达水平。结果与 IR 组相比齐墩果酸组血清肝酶水平及肝组织中 MDA 水平显著降低,而 SOD 和 GSH 活性则显著升高;与 IR组相比齐墩果酸组肝组织病理形态学损伤明显减轻;缺血再灌注后可见明显的肝实质细胞凋亡现象,齐墩果酸组凋亡细胞较 IR 组显著下降,且 caspase3和 p53蛋白表达亦减少。结论齐墩果酸预处理可减轻肝脏缺血再灌注损伤,可能与抑制 caspase3和 p53蛋白表达从而抑制细胞凋亡有关。

목적:관찰제돈과산(OA)예처리대간장결혈재관주손상중세포조망적영향,탐토제돈과산예처리대간장보호작용적궤제。방법웅성 SD 대서40지,수궤분위가수술조(SH 조)、결혈재관주조(IR 조)、0.5%최갑기섬유소납조(CM 조)화제돈과산예처리조(제돈과산조),제돈과산조이100 mg/kg 적제돈과산혼현액,SH 화 IR 조이상동용적적수,CM 조이상동용적적0.5%CMC-Na 분별매일관위1차,련속7 d。제8천건립70%간장결혈재관주모형,간장결혈60 min 후행재관주3 h。검측대서혈청병안산안기전이매(ALT)、천동안산안기전이매(AST)화LDH 수평;검측간조직내병이철(MDA)、초양화물기화매(SOD)、곡광감태(GSH)수평변화;통과 HE 염색관찰간조직형태,이용 Tunel 염색검측간조직조망상태;Western blot 검측간장조직 caspase3화 p53단백표체수평。결과여 IR 조상비제돈과산조혈청간매수평급간조직중 MDA 수평현저강저,이 SOD 화 GSH 활성칙현저승고;여 IR조상비제돈과산조간조직병리형태학손상명현감경;결혈재관주후가견명현적간실질세포조망현상,제돈과산조조망세포교 IR 조현저하강,차 caspase3화 p53단백표체역감소。결론제돈과산예처리가감경간장결혈재관주손상,가능여억제 caspase3화 p53단백표체종이억제세포조망유관。
Objective To investigate the effect of oleanolic acid on cell apoptosis in rat livers with ischemia-reperfusion Injury.Methods Forty healthy Wistar rats were randomly divided into 4 groups including sham-operated group(SH)which was reperfused with water,Ischemia-Reperfusion group(IR)which was reperfused with water,0. 5% sodium carboxymethyl cellulose group(CM)which was reperfused with 0.5% sodium carboxymethyl cellulose and oleanolic acid group(OA)which was reperfused with oleanolic acid.The four groups were all reperfused seven days.At the eighth day,70% liver ischemia reperfusion models were established,and after the liver ischemia 60 min, reperfused 3 h.The serum ALT,AST and LDH were detected as liver damage markers.MDA,SOD and GSH in liver were measured.The expressions of caspase3 and p53 were detected by western blot.The apoptosis was measured by Tunel.The histological observation was carried out with hematoxylin and eosin staining.Results Compared with the IR group,the activity of plasma enzyme and the concentration of MDA in hepatic tissue in the OA group were markedly reduced,while the activity of SOD and GSH were enhanced.There were obvious hepatocellular apoptosis after reperfusion and the apoptptic index in the OA group was significantly lower than of the IR group.The expres-sions of caspase3 and p53 were significant lower in the OA group than of the IR group.Conclusion Oleanolic acid could reduce hepatocellular apoptosis after reperfusion,and thus produce a protective effect on hepatic ischemia-reper-fusion injury.
