目的:探讨适宜的人凝血因子Ⅷ制剂中蛋白含量测定方法。方法分别应用《中国药典》三部(2010年版)中凯氏定氮法和 Lowry 法测定人凝血因子Ⅷ制剂中的蛋白含量,比较两种方法的稳定性,并以甘氨酸为干扰物质,考察甘氨酸对 Lowry 法的干扰作用。结果两种方法在各自适合的测定范围内,凯氏定氮法的准确度优于Lowry 法,在测定人凝血因子Ⅷ制剂中的蛋白含量时,凯氏定氮法的回收率 RSD 明显高于 Lowry 法。结论Low-ry 法易操作,结果稳定,灵敏度较高,较凯氏定氮法更适合于人凝血因子Ⅷ制剂中的蛋白含量测定。但甘氨酸可干扰 Lowry 法的测定结果,随着甘氨酸含量的增加,干扰作用明显增强。

목적:탐토괄의적인응혈인자Ⅷ제제중단백함량측정방법。방법분별응용《중국약전》삼부(2010년판)중개씨정담법화 Lowry 법측정인응혈인자Ⅷ제제중적단백함량,비교량충방법적은정성,병이감안산위간우물질,고찰감안산대 Lowry 법적간우작용。결과량충방법재각자괄합적측정범위내,개씨정담법적준학도우우Lowry 법,재측정인응혈인자Ⅷ제제중적단백함량시,개씨정담법적회수솔 RSD 명현고우 Lowry 법。결론Low-ry 법역조작,결과은정,령민도교고,교개씨정담법경괄합우인응혈인자Ⅷ제제중적단백함량측정。단감안산가간우 Lowry 법적측정결과,수착감안산함량적증가,간우작용명현증강。
Objective To investigate the suitable methods for the protein determination coagulation factor Ⅷ. Methods The protein content in the coagulation factor Ⅷ was determined by Kjeldahl′s assay and Lowry′s assay de-scribed in Chinese Pharmacopeia(VolumeⅢ,2010 edition),using glycine as an interfering substance,observe the in-terference effects of glycine on Lowry method.Results In their suitable determination range,the accuracy of Kjel-dahl′s assay was better than that of Lowry′s assay.The recovery rates of Kjeldahl′s assay was significantly higher than that of Lowry′s assay in the determination of protein content.Conclusion Lowry′s assay was easy to operate, the results was stable and high sensitivity.Compared with the Kjeldahl′s assay ,Lowry′s assay was more suitable for determining protein content in coagulation factor Ⅷ.The determination results were interfered by glycine.With the increase of glycine content,the effect enhanced obviously.
