目的:将DNA直接扩增技术引入案件现场不同的接触性检材的检验中,构建有效的接触性DNA的生物检材的检验方法。方法采用剪取、擦拭等方法对手套、烟蒂、衣服、饮料瓶、吸管及残缺指纹等检材进行取样,采用DNA直接扩增技术获得短串联康复序列(short tandem repeat ,STR)分型。结果 DNA直接扩增技术在5种不同检材中检出的基因型均为单一个体来源,其对手套、烟蒂、衣服、饮料瓶和吸管、残缺指纹检材的成功率分别为90%、100%、70%、90%和60%,平均成功率在70%以上。分别对5种检材多处取点样品进行平行扩增,手套、烟蒂检材3个点位成功率均在90%以上;饮料瓶、吸管3个点位的成功率达80%~90%;衣服3个点位的成功率在60%~70%;残缺指纹3个点位的成功率在50%~60%。结论 DNA直接扩增技术对接触性DNA检材检验具有较高的有效应,其结果可靠,重复性强。

목적:장DNA직접확증기술인입안건현장불동적접촉성검재적검험중,구건유효적접촉성DNA적생물검재적검험방법。방법채용전취、찰식등방법대수투、연체、의복、음료병、흡관급잔결지문등검재진행취양,채용DNA직접확증기술획득단천련강복서렬(short tandem repeat ,STR)분형。결과 DNA직접확증기술재5충불동검재중검출적기인형균위단일개체래원,기대수투、연체、의복、음료병화흡관、잔결지문검재적성공솔분별위90%、100%、70%、90%화60%,평균성공솔재70%이상。분별대5충검재다처취점양품진행평행확증,수투、연체검재3개점위성공솔균재90%이상;음료병、흡관3개점위적성공솔체80%~90%;의복3개점위적성공솔재60%~70%;잔결지문3개점위적성공솔재50%~60%。결론 DNA직접확증기술대접촉성DNA검재검험구유교고적유효응,기결과가고,중복성강。
Objective The effective protocal was constructed for DNA of different contact biological materials at the crime scene using direct amplification of DNA .Methods The samples were selected from contact materials such as gloves ,cigarette end ,clothes ,beverage bottle ,suction pipe and incomplete fingerprint by scissors and wi-ping .Then STR type was obtained using direct PCR .Results Genotype derived from five contact materials was the origin of a single individual by direct amplification of DNA .The successful rates were 90% ,100% ,70% ,90%and 60% in gloves ,cigarette end ,clothes ,beverage bottle ,suction pipe and incomplete fingerprint ,respectively . And the average successful rate was more than 70% .Parallel amplification was used for five contact materials by multidraw .The successful rate was more than 90% from 3 points in gloves and cigarette end ,80% ~90% in bev-erage bottle and suction pipe ,60% ~70% in clothes ,and 50% ~60% in incomplete fingerprint .Conclusion The direct amplification of DNA is efficient for DNA of contact materials .The results are reliable and good repeatabili-ty .
