目的:比较研究Aβ在两种AD转基因小鼠模型脑杏仁核分布的差异。方法:采用18月龄雄性APP/PSl双转基因(2×Tg-AD)小鼠与同龄同性别APP/PSl/tau三转基因(3×Tg-AD)小鼠,分别进行6E10单克隆抗体免疫组化染色等方法显示Aβ阳性神经元及斑块,观察其分布与形态等的差异,图像分析系统定量比较其量的变化。结果:在杏仁核2×Tg-AD组Aβ阳性产物主要位于细胞外成为细胞外Aβ( eAβ),形成大量的Aβ阳性斑,Aβ阳性神经元少;而3×Tg-AD组Aβ阳性产物主要位于神经元细胞内,成为细胞内Aβ( iAβ),但Aβ阳性斑少见。结论:2×Tg-AD组与3×Tg-AD组Aβ阳性产物在杏仁核分布的差异可能反映了两种AD小鼠模型神经病理等改变的不同。

목적:비교연구Aβ재량충AD전기인소서모형뇌행인핵분포적차이。방법:채용18월령웅성APP/PSl쌍전기인(2×Tg-AD)소서여동령동성별APP/PSl/tau삼전기인(3×Tg-AD)소서,분별진행6E10단극륭항체면역조화염색등방법현시Aβ양성신경원급반괴,관찰기분포여형태등적차이,도상분석계통정량비교기량적변화。결과:재행인핵2×Tg-AD조Aβ양성산물주요위우세포외성위세포외Aβ( eAβ),형성대량적Aβ양성반,Aβ양성신경원소;이3×Tg-AD조Aβ양성산물주요위우신경원세포내,성위세포내Aβ( iAβ),단Aβ양성반소견。결론:2×Tg-AD조여3×Tg-AD조Aβ양성산물재행인핵분포적차이가능반영료량충AD소서모형신경병리등개변적불동。
Objective:To compare the distribution and morphological features of Aβimmunoreactive products in the amygdale of two transgenic mice models of Alzheimer's disease.Methods:18-month-old APP/PSl ( 2 ×Tg-AD) and the age-matched male mice of APP/PSl/tau(3 ×Tg-AD) were used in this experiment.6E10 immunohis-tochemical staining was used to show the distribution and morphological features of Aβimmunoreactive products, and quantitative analysis were made on the difference of the products by an image analysis system.Results:The deposition of Aβin the amygdala of 2 ×Tg-AD was mainly seen as extracellular Aβ( eAβ) , and formed a lot of positive plaques;the positive products were distributed as intracellularly Aβ( iAβ) for the 3 ×Tg-AD, but the Aβpositive plaques were rarely seen.Conclusion:The difference of Aβpositive products distribution between the 2 ×Tg-AD and 3 ×Tg-AD is likely to be related to the difference of neuropathological changes of these two AD models.
