目的:探讨白细胞介素-17F基因多态性与儿童支气管哮喘的相关性,为支气管哮喘的易感基因筛选提供可能的理论依据。方法取本院门诊随诊的60例哮喘患儿设为哮喘组,60例健康儿童设为对照组,两组儿童均于清晨空腹留取外周静脉血2 ml,提取全基因组 DNA,应用聚合酶链反应及克隆、基因测序方法分析 IL-17F基因序列rs763780(H161R)基因多态性。结果 IL-17F基因序列 rs763780(H161R)基因型在两组内均未检测到突变型 CC, rs763780(H161R)基因型(TC/TT)在两组间内的分布有明显的差异(χ2=5.71,P<0.05)。TC基因型 OR和95%可信区间(CI)为2.09(1.12-3.90),TT基因型 OR和95%CI为0.76(0.60-0.95)。结论 IL-17F基因多态性 rs763780(H 161 R)基因型分布(TC/TT)在两组间存在差异,其多态性可能与哮喘相关,两组内均未发现基因型 CC在,基因型TC可能是哮喘发病的危险因素。

Objective To explore the correlation between IL-17F gene polymorphism and children asthma,providing some theoretical basis for pathophysiology and identification of susceptibility to asthma.Methods 60 Follow-up chil-dren with asthma in clinic were enrolled randomly to be asthma group,sixty healthy children were set as control group. 2 ml peripheral vein blood was drawn from each child with empty stomach in early morning.Each blood sample was ex-tracted genomic DNA,Polymerase chain reaction (PCR),gene clone and DNA sequencing were used to analyse the IL-17F polymorphisms (rs763780).Results The H161R homozygote (CC )is not found in both asthma group and health group.Significant deviations from the genotypes (TC/TT)of rs763780 were found in the asthma and health group.(χ2=5.71 P=0.017<0.05).The OR for asthma was 2.09 (95% CI,1.12-3.90)for the H161R mutant type(TC),the OR for asthma was 0.76 (95% CI,0.60-0.95)for the H161R wild type(TT).Conclusion Significant deviations from the genotypes (TC/TT)of IL-17F rs763780 were found between the asthma and control groups,the polymorphisms may be associated with asthma.The H161R homozygote (CC)is not found in both groups,The H161R mutant type (TC)may be the risk of asthma.
