目的:探索在位子宫内膜间质细胞体外分离、纯化、培养的方法,从而建立研究子宫内膜异位症(EMs)的体外细胞模型。方法:选择因子宫肌瘤或子宫腺肌症行子宫(次)全切除术的病例38例,术中取其子宫内膜,通过酶解、过滤、贴壁纯化等技术,分离培养子宫内膜间质细胞并进行传代。结果:36例标本获得成功,分离纯化得到的子宫内膜间质细胞经角蛋白、波形蛋白染色证实纯度达95%以上,活性较强,可以有限传代,传代培养的细胞形态特征与原代细胞一致。结论:该方法可以高效简便地分离在位子宫内膜间质细胞,得到的间质细胞产量及纯度高,可以作为 EMs 的体外细胞模型之一。

목적:탐색재위자궁내막간질세포체외분리、순화、배양적방법,종이건립연구자궁내막이위증(EMs)적체외세포모형。방법:선택인자궁기류혹자궁선기증행자궁(차)전절제술적병례38례,술중취기자궁내막,통과매해、과려、첩벽순화등기술,분리배양자궁내막간질세포병진행전대。결과:36례표본획득성공,분리순화득도적자궁내막간질세포경각단백、파형단백염색증실순도체95%이상,활성교강,가이유한전대,전대배양적세포형태특정여원대세포일치。결론:해방법가이고효간편지분리재위자궁내막간질세포,득도적간질세포산량급순도고,가이작위 EMs 적체외세포모형지일。
Objective:Exploring a method to separate、purify and culture eutopic endometrial stromal cells, in order to establish an in vitro cell model for researching the disease of endometriosis. Methods: Selecting 38 patients whose hysterectomy will be performed for myoma or adenomyoma, to obtain the endometrium when the operation is on. Getting primary cultures of stromal cells from endometrium by techniques of enzymolysis、filtration、adherence and purification etal. Results: The endometrial stromal cells were successfully cultured in 36 of 38 samples. The purities of stromal cells were 95%.Stromal cells could be transferred into several passage and the morphologic characteristics would not be changed. Conclusions: It is effective and convenient to get eutopic endometrial stromal cells with high yield and purity by our method. It can be one of cell models in vitro for endometriosis.
