研究用于甘油二酯制备的国产脂肪酶的固定化方法及酶解工艺。方法:以12种国产树脂为载体,采用物理吸附法对脂肪酶L04进行固定化的载体筛选实验,并对胜出者进行酶解条件优化。结果:采用叔胺基树脂固定化脂肪酶的酶活最高,ESQ-1-X树脂与酶的质量比为15:1时,固载效果最佳,采用0.1g/g(酶/树脂)固载酶用量,酶解时间20h可得到56.1%甘油二酯得率,固载酶循环使用6次仍保持产品得率基本不变。

The immobilization of lipase and enzyme hydrolyzation technology were studied for preparing of the diglyceride in this paper. Lipase (L04) was immobilized by 12 kinds of resins, the excellent resin was selected as subject to optimize further for preparing diglyceride. The results shown that the resin modified, the lipase (L04) was immobilized onto the resin containing tertiary amino groups (ESQ-1-X) and the mass ratio of ESQ-1-X to lipase (L04) was found to be 15:1, possessed excellent lipase activity. 56.1% of the diglyceride in the reaction system was acquired by adding 0.1g/g (lipase/resin) of the resin modified and the reaction time was 20h, the yield of diglyceride remained fairly constant after having recycled for 6 times.
