蒺藜是一种常用的药用植物,含有多种有效成分,其中总皂苷和总黄酮是其主要成分,总黄酮的含量是总皂苷的1.5倍,因此对蒺藜中总黄酮的提取工艺进行了研究。实验中采用正交设计法对蒺藜中总黄酮的最佳提取工艺进行优化。通过单因素实验得到的初步结果首先确定了四因素,分别为:液固比、提取时间、提取次数和乙醇浓度,同时确定每个因素的三个水平。以总黄酮的含量为考察指标,根据L9(34)正交实验法进行9次实验。研究结果表明:液固比25:1、提取时间10h、提取次数4次、乙醇浓度70%为蒺藜中总黄酮的最佳回流提取工艺。经过结果分析得出:四因素中液固比是实验结果最大影响因素,提取时间对结果也有显著影响,提取次数和乙醇浓度对结果的影响较小。后续验证研究表明该最佳回流提取工艺条件稳定,提取效率较好。

Tribulus is a common medicinal plant that contains a variety of effective ingredients including the total saponins and the total flavonoids are the main ones, and the content of the total flavonoids is 1.5 times more than the saponins. So the extracting technology of the flavonoids from the Tribulus is being studied. We optimize the best extracting technology by the orthogonal design. First depending on the preliminary results which get by the single factor experiments, we choose the four factors, such as the liquid-solid ratio, extraction time, extraction time and concentration of the ethanol, and in the same time we determine the three levels of each factors, and regard the content of total flavonoids as the index. Then conducting nine experiments on the basis of the L9(34) orthogonal design. The results of the studies show that the ratio of liquid to solid and extraction time are 25:1 and 10h, respective. And when tribulus is extracted for 4 times by 70% ethanol, then the total flavonoids has the highest yield. After the analysis of the results, we can draw obtain a conclusion: among the four factors designed in the experiment, the liquid-solid ratio has the greatest impact on the experimental results, the extraction time has a significant influence, and the number of extraction and ethanol concentration have little effect. The later studies show that it is the best reflux extraction process and the extraction efficiency is good.
