目的:比较卵巢癌SK-OV-3细胞在2D和3D培养系统中的生长特性。方法将卵巢癌细胞株SK-OV-3分别采用2D和3D培养系统进行培养,观察细胞生长形态。采用CCK-8法测定细胞生长,Brdu法检测细胞增殖,流式细胞术检测细胞周期分布。结果 SK-OV-3细胞在2D培养系统中呈单层贴壁生长;在3D培养系统中形成多细胞球样体(muti-cellular spheroid,MCS),生长速度较2D培养系统慢。在2D培养系统中SK-OV-3细胞的S/G2-M期细胞比例为(53.7±5.8)%,明显高于在3D培养系统中S/G2-M期细胞比例[(40.9±2.0)%,P<0.05]。结论不同的培养方式对卵巢癌细胞的生长具有较大的影响。MCS三维培养系统能更好地模拟体内肿瘤细胞的生长状况,是卵巢癌研究的良好平台。

Objective To compare the growth characteristics of ovarian cancer cells in 2D or 3D culture system. Methods Human ovarian cancer cells of SK-OV-3 were maintained under 2D or 3D culture systems. Cells morphology was observed by the inverted microscope and the growth rate was detected by CCK-8 test re-spectively. The cells ’ proliferation and cycle distribution were analyzed by Brdu assay and flow cytometry (FCM)respectively. Results Maintained under 2D culture system,SK-OV-3 cells grew in monolayer;but under 3D culture system,cells formed multi-cellular spheroids (MCS)with a bit slower growth and proliferation rates. The proportion of S/G2-M phase of cells cultured in 2D system was significantly more than those in 3D system [(53. 7 ± 5. 8)% vs. (40. 9 ± 2. 0)%,P<0. 05]. Conclusions The characteristics of ovarian canc-er cells in 2D and 3D culture systems are different. MCS culture system can better mimic the in vivo growth en-vironment of tumors and could be a better model for ovarian cancer research.
