以钙离子荧光指示剂Fluo-3-AM作为神经细胞内Ca2+荧光探针,用微孔板荧光检测仪测量神经细胞内游离Ca2+与荧光探针Fluo-3-AM结合的荧光强度,并实时检测此荧光强度在细胞外加不同浓度腺苷诱导下的变化情况。实验发现,各种浓度的腺苷都能诱导神经细胞Ca2+浓度升高,且其影响程度随腺苷浓度的增大而增强。这一结果对探究腺苷通过作用神经系统起止痛作用的机理具有重要意义。
以鈣離子熒光指示劑Fluo-3-AM作為神經細胞內Ca2+熒光探針,用微孔闆熒光檢測儀測量神經細胞內遊離Ca2+與熒光探針Fluo-3-AM結閤的熒光彊度,併實時檢測此熒光彊度在細胞外加不同濃度腺苷誘導下的變化情況。實驗髮現,各種濃度的腺苷都能誘導神經細胞Ca2+濃度升高,且其影響程度隨腺苷濃度的增大而增彊。這一結果對探究腺苷通過作用神經繫統起止痛作用的機理具有重要意義。

Calcium ion fluorescent tracers of Fluo-3-AM is used as the Ca2 +fluorescent probe in neurons and the fluorescence intensity combining the intracellular free Ca2+ and the fluorescent probe Fluo-3-AM in neurons is determined by micro-well plate fluorescence detection. And then the fluorescence intensity of the neurons in real-time is determined when the neurons are induced by different concentrations of adenosine. It’ s discovered that the concentration of Ca2+ in neurons is going up slowly when the neurons are induced by adenosine. The influencing degree increases with the increase of the concentration of adenosine. This experiment results are quite important for people to explore the analgesic mechanism of adenosine which affects the function of the nervous system.
