目的：通过体内体外实验，观察白藜芦醇（resveratrol，Res）对肝癌细胞Bel-7402的增殖抑制作用，并对其抑癌的可能效应机制进行分析。方法实验中设4个Res药物组，终浓度分别为12.5、25、50、100μmol/L，同时设置不含Res药物的对照组，采用MTT法测试Res对体外培养肝癌Bel-7402细胞的增殖抑制作用，反转录PCR（reverse transcription-PCR，RT-PCR）检测Bcl-2 mRNA的表达，Western Blot检测Bcl-2蛋白的表达，ELISA测定Res对荷瘤小鼠细胞因子水平的影响。结果与对照组相比，Res可以呈剂量和时间依赖性方式抑制肝癌Bel-7402细胞的增殖和Bcl-2 mRNA及其蛋白的表达（P＜0.05）。同时Res能明显抑制荷瘤小鼠肿瘤的生长，并能明显升高荷瘤小鼠体内细胞因子IL-2、IL-6、IL-12和TNF-α的水平（P＜0.05）。结论 Res体内与体外对肝癌细胞Bel-7402均有明显的增殖抑制活性，其抗肿瘤的效应机制可能与其下调Bcl-2的表达和提高细胞因子IL-2、IL-6、IL-12及TNF-α的水平有关。
목적：통과체내체외실험，관찰백려호순（resveratrol，Res）대간암세포Bel-7402적증식억제작용，병대기억암적가능효응궤제진행분석。방법실험중설4개Res약물조，종농도분별위12.5、25、50、100μmol/L，동시설치불함Res약물적대조조，채용MTT법측시Res대체외배양간암Bel-7402세포적증식억제작용，반전록PCR（reverse transcription-PCR，RT-PCR）검측Bcl-2 mRNA적표체，Western Blot검측Bcl-2단백적표체，ELISA측정Res대하류소서세포인자수평적영향。결과여대조조상비，Res가이정제량화시간의뢰성방식억제간암Bel-7402세포적증식화Bcl-2 mRNA급기단백적표체（P＜0.05）。동시Res능명현억제하류소서종류적생장，병능명현승고하류소서체내세포인자IL-2、IL-6、IL-12화TNF-α적수평（P＜0.05）。결론 Res체내여체외대간암세포Bel-7402균유명현적증식억제활성，기항종류적효응궤제가능여기하조Bcl-2적표체화제고세포인자IL-2、IL-6、IL-12급TNF-α적수평유관。
Objective To explore the inhibition effect and the possible mechanism of resveratrol on human hepatocellular carcinoma cell line Bel-7402 both in vitro and vivo.Methods Four Res drugs in the experiment group,the final concentrations were 12.5,25,50,100μmol/L,the control group at the same time set not containing Res drugs,MTT assay was used to measure the inhibition of resveratrol on Bel-7402.The expression of Bcl-2 was detected by RT-PCR and Western Blot.The levels of IL-2,IL-6,IL-12 and TNF-αwere detected by ELISA.Results Resveratrol inhibited Bel-7402 cell proliferation in dose and time manner,and influenced the expression of Bcl-2 mRNA and protein.At the same time,resveratrol inhibited the growth of tumor and improved the levels of IL-2,IL-6,IL-12 and TNF-α.Conclusion Resveratrol could inhibit Bel-7402 cell proliferation both in vitro and vivo, the possible mechanism may be that resveratrol could low down the expression of Bcl-2 and improve the levels of IL-2,IL-6,IL-12 and TNF-α.
