目的探讨黄芪总皂苷对急性心肌梗死( AMI)大鼠缺血心肌组织血管内皮生长因子( VEGF)蛋白表达水平的影响。方法雄性Wistar大鼠24只,建立AMI模型,术后24 h存活大鼠随机分为药物组(n=8)、对照组(n=8),另设假手术组(n=8)。药物组给予黄芪总皂苷水溶液2.5 mg/(kg· d)腹腔注射,对照组及假手术组给予等体积生理盐水,4周处死大鼠。心肌组织取材,Ⅷ因子染色计数微血管数( MVC)及微血管密度( MVD),免疫组化法观察缺血心肌VEGF蛋白表达水平。结果药物组及对照组大鼠缺血心肌MVC、MVD均高于假手术组,且药物组MVC、MVD高于对照组(P<0.05);药物组及对照组大鼠缺血心肌VEGF蛋白表达均高于假手术组(P<0.01),且药物组高于对照组(P<0.01)。结论黄芪总皂苷促进大鼠缺血心肌血管新生,同时可上调缺血心肌组织VEGF蛋白表达水平。

목적탐토황기총조감대급성심기경사( AMI)대서결혈심기조직혈관내피생장인자( VEGF)단백표체수평적영향。방법웅성Wistar대서24지,건립AMI모형,술후24 h존활대서수궤분위약물조(n=8)、대조조(n=8),령설가수술조(n=8)。약물조급여황기총조감수용액2.5 mg/(kg· d)복강주사,대조조급가수술조급여등체적생리염수,4주처사대서。심기조직취재,Ⅷ인자염색계수미혈관수( MVC)급미혈관밀도( MVD),면역조화법관찰결혈심기VEGF단백표체수평。결과약물조급대조조대서결혈심기MVC、MVD균고우가수술조,차약물조MVC、MVD고우대조조(P<0.05);약물조급대조조대서결혈심기VEGF단백표체균고우가수술조(P<0.01),차약물조고우대조조(P<0.01)。결론황기총조감촉진대서결혈심기혈관신생,동시가상조결혈심기조직VEGF단백표체수평。
Objective To study the effect of Astragalus on the level of vascular endothelial growth factor ( VEGF) in ischemic myocardium of rats.Methods The acute myocardial infarction ( AMI) models were established in twenty-four male Wistar rats.Twenty-four hours after procedure, the surviving rats were randomly divided into drug group ( n=8 ) , control group ( n=8 ) , and sham-operated group (n =8).Rats in the drug group were injected with Astragalus [2.5 mg/(kg· d)] Intraperitoneally.The control group and the sham-operated group were received same volume saline intraperitoneally.Four weeks later, the rats were sacrificed and the myocardial tissue were collected. The number of miniature blood vessel ( MVC) and microvascular density ( MVD) was counted according to FⅧ staining technique, and the expression of VEGF was detected by immunohistochemistry technique.Results The number of MVC and MVD in drug group and control group was higher than that in sham-operated group, and the drug group was higher than the control group (P<0.05); higher expression of VEGF was shown in drug group and control group than sham-operated group (P<0.01), while the expression in drug group was higher than control group (P<0.01).Conclusion Astragalus can increase the level of VEGF and promote angiogenesis in ischemic myocardium of rats.
