目的探讨甘草酸二铵( DG)对百草枯中毒( PQ)致急性肺损伤( ALI)大鼠肺Toll样受体4(TLR-4) mRNA、NF-κB mRNA表达及对血清白细胞介素-6(IL-6)、白细胞介素-17(IL-17)浓度的影响。方法将50只SD大鼠采用随机数字表法分为PQ组(n=20)、DG治疗组(n=20)和正常对照组(S组,n=10),PQ组和DG治疗组予百草枯100 mg/kg灌胃1次,DG组于灌胃后立即腹腔注射DG 50 mg/kg(1次/d),S组和PQ组注射等剂量的生理盐水,观察至48 h。取肺组织检测肺湿干质量比( W/D);采用RT-PCR法检测肺组织TLR-4 mRNA、NF-κB mRNA的表达情况;ELISA法测定血清IL-6、IL-17浓度。另选择健康SD大鼠50只,分组及各组处置方法同上,观察其7 d内死亡率。结果与S组比较,PQ组W/D、TLR-4 mRNA、NF-κB mRNA表达均明显升高(P<0.01),血清IL-6、IL-17亦升高(P<0.01);DG组干预后,W/D降低, TLR-4 mRNA、NF-kB mRN、AIL-6及IL-17有所降低,与PQ组比较差异有统计学意义( P<0.01或P<0.05)。 PQ组大鼠7 d死亡18只(死亡率90%),DG组死亡8只(死亡率40%),NS组无死亡(死亡率0)。结论甘草酸二铵可抑制PQ致ALI大鼠肺组织TLR-4 mRNA、NF-κB mRNA及血清IL-6、IL-17浓度的过度表达,从而减轻肺损伤程度。

목적탐토감초산이안( DG)대백초고중독( PQ)치급성폐손상( ALI)대서폐Toll양수체4(TLR-4) mRNA、NF-κB mRNA표체급대혈청백세포개소-6(IL-6)、백세포개소-17(IL-17)농도적영향。방법장50지SD대서채용수궤수자표법분위PQ조(n=20)、DG치료조(n=20)화정상대조조(S조,n=10),PQ조화DG치료조여백초고100 mg/kg관위1차,DG조우관위후립즉복강주사DG 50 mg/kg(1차/d),S조화PQ조주사등제량적생리염수,관찰지48 h。취폐조직검측폐습간질량비( W/D);채용RT-PCR법검측폐조직TLR-4 mRNA、NF-κB mRNA적표체정황;ELISA법측정혈청IL-6、IL-17농도。령선택건강SD대서50지,분조급각조처치방법동상,관찰기7 d내사망솔。결과여S조비교,PQ조W/D、TLR-4 mRNA、NF-κB mRNA표체균명현승고(P<0.01),혈청IL-6、IL-17역승고(P<0.01);DG조간예후,W/D강저, TLR-4 mRNA、NF-kB mRN、AIL-6급IL-17유소강저,여PQ조비교차이유통계학의의( P<0.01혹P<0.05)。 PQ조대서7 d사망18지(사망솔90%),DG조사망8지(사망솔40%),NS조무사망(사망솔0)。결론감초산이안가억제PQ치ALI대서폐조직TLR-4 mRNA、NF-κB mRNA급혈청IL-6、IL-17농도적과도표체,종이감경폐손상정도。
Objective To evaluate the effect of diammonium glycyrrhizinate ( DG ) on paraquat-induced acute lung injury in rats.Methods Fifty SD rats were randomly divided into control group ( NS ) , PQ group and DG group.The rats in PQ group and DG group were intragastriced administration with paraquat (100 mg/kg) .The rats in DG group were intragastriced administration with DG (50 mg/kg) once per day after paraquat injection.Pulmonary lung wet to dry weight ratio was determined;the lung tissue HE staining was used to evaluate the pulmonary tissue injury;to detect the expression of lung tissue TLR -4 mRNA and NF -κB mRNA RT -PCR method was used.The concentration of IL-6 and IL-17 in serum were determined by ELISA.Results Compared with the control group, the lung tissue's wet/dry weight ratio, expression of lung tissue TLR-4 mRNA, NF-κB mRNA, IL-6 and IL -17 in serum increased significantly and comparison between groups showed statistical significance (P<0.01 or P<0.05).7 day mortality was 90% in PQ group, 40% in DG group, and no death in NS group.Conclusion DG can attenuate the acute lung injury induced by paraquat poisoning in rats, the mechanism may be related to the decrease of TLR -4 and NF -κB mRNA expression.
