目的:观察血必净注射液对脓毒症大鼠肠道炎症因子表达的变化的影响作用,探索其在脓毒症发病中的作用机制和意义。方法:将72只SD雄性大鼠随机分成对照组、模型组、治疗组,每组各24只。模型组和治疗组用CLP法制备脓毒症大鼠模型。治疗组制模前3d起给予血必净2ml/只腹腔注射,1次/12h ,共5d。其他两组注射相同体积生理盐水。各时间点经腹主动脉采血,距回盲部5cm处取回肠3cm ,ELISA测定血浆血小板活化因子(PAF),回肠匀浆中肿瘤坏死因子-α(TNF-α)、PCR测定组织血小板活化因子受体(Pafr)mRNA 表达水平。结果:模型组各时间点PAF表达明显高于对照组(P<0.05),Pafr表达明显低于对照组(P<0.05),12h、24h、48h TNF-α表达明显高于对照组(P<0.05),血必净可明显抑制这种变化(P<0.05)。结论:脓毒症可致大鼠肠 TNF-α、PAF分泌增加,Pafr基因表达下降,血必净对肠道的保护作用与抑制PAF的生成和抑制PAF与Pafr的结合有关。
目的:觀察血必淨註射液對膿毒癥大鼠腸道炎癥因子錶達的變化的影響作用,探索其在膿毒癥髮病中的作用機製和意義。方法:將72隻SD雄性大鼠隨機分成對照組、模型組、治療組,每組各24隻。模型組和治療組用CLP法製備膿毒癥大鼠模型。治療組製模前3d起給予血必淨2ml/隻腹腔註射,1次/12h ,共5d。其他兩組註射相同體積生理鹽水。各時間點經腹主動脈採血,距迴盲部5cm處取迴腸3cm ,ELISA測定血漿血小闆活化因子(PAF),迴腸勻漿中腫瘤壞死因子-α(TNF-α)、PCR測定組織血小闆活化因子受體(Pafr)mRNA 錶達水平。結果:模型組各時間點PAF錶達明顯高于對照組(P<0.05),Pafr錶達明顯低于對照組(P<0.05),12h、24h、48h TNF-α錶達明顯高于對照組(P<0.05),血必淨可明顯抑製這種變化(P<0.05)。結論:膿毒癥可緻大鼠腸 TNF-α、PAF分泌增加,Pafr基因錶達下降,血必淨對腸道的保護作用與抑製PAF的生成和抑製PAF與Pafr的結閤有關。
목적:관찰혈필정주사액대농독증대서장도염증인자표체적변화적영향작용,탐색기재농독증발병중적작용궤제화의의。방법:장72지SD웅성대서수궤분성대조조、모형조、치료조,매조각24지。모형조화치료조용CLP법제비농독증대서모형。치료조제모전3d기급여혈필정2ml/지복강주사,1차/12h ,공5d。기타량조주사상동체적생리염수。각시간점경복주동맥채혈,거회맹부5cm처취회장3cm ,ELISA측정혈장혈소판활화인자(PAF),회장균장중종류배사인자-α(TNF-α)、PCR측정조직혈소판활화인자수체(Pafr)mRNA 표체수평。결과:모형조각시간점PAF표체명현고우대조조(P<0.05),Pafr표체명현저우대조조(P<0.05),12h、24h、48h TNF-α표체명현고우대조조(P<0.05),혈필정가명현억제저충변화(P<0.05)。결론:농독증가치대서장 TNF-α、PAF분비증가,Pafr기인표체하강,혈필정대장도적보호작용여억제PAF적생성화억제PAF여Pafr적결합유관。
Objective:To investigate the intervention of Xuebijing injection on expression of inflammatory cytokines in the septic rats’intestines .Methods:72 male SD rats were divided into 3 groups :control group ,model group and treat-ment group .The septic model was prepared by cecal ligation and puncture (CLP) .The model and the treatment group took CLP .3 days before modeling ,the rats of the treatment group were beginning to be injected with Xuebijing in the abdomen at a dosage of 2ml/rat ,once for 12 hours and 5 days in total ,while rats in the other 2 groups were treated with the same dosage of saline solution .Rats in each group at each time point were sacrificed after blood collection via aorta . The blood was used to measure the level of PAF by ELISA .And 2 pieces of 3cm ileum from the distance of 5cm to ileo-cecum were collected for measuring the level of TNF-α by ELISA and the expression of Pafr .Results:From 6h time point to 48 h time point ,the level of PAF and Pafr in model group changed remarkably compared with the control group (P<0 .05) ,from 12h time point to 48h time point ,the level of TNF-αin model group increased remarkably com-pared with the control group (P<0 .05) .The changes were restrained remarkably after injection of xuebijing .Conclu-sion:In septic intestines ,the level of TNF-αand PAF increased ,the expression of Pafr decreased .Xuebijing realizes its protective effect by suppressing the expression of inflammatory cytokines or the combination of PAF and Pafr .
