目的:通过观察2型糖尿病(T2DM)大鼠脂联素(APN)及心肌脂联素受体1的表达情况,探讨其在T2DM心肌病发病过程中的作用。方法建立T2DM大鼠动物模型。采用双抗体夹心酶联免疫法检测血清及心肌APN水平,并采用免疫组化的方法测定心肌脂联素受体1(AdipoR1)的蛋白表达。结果与对照组比较,T2DM组血清和心肌APN水平下降,心肌AdipoR1蛋白的表达水平降低,差异均有统计学意义(P<0.05)。相关性分析显示,T2DM组大鼠血清APN与血清胰岛素、稳态模型胰岛素抵抗指数呈负相关(r=-0.613,P<0.05;r=-0.637,P<0.05),心肌AdipoR1蛋白表达量与心肌APN呈正相关(r=0.89,P<0.05),与HOMA稳态模型(HOMA-IR)及空腹胰岛素水平(FINS)呈负相关(r=-0.697,P<0.05;r=-0.593,P<0.05)。结论糖尿病心肌病大鼠血清及心肌APN水平下降,心肌AdipoR1蛋白表达下降,胰岛素抵抗可能在糖尿病心肌病的发病过程中起到作用。

목적:통과관찰2형당뇨병(T2DM)대서지련소(APN)급심기지련소수체1적표체정황,탐토기재T2DM심기병발병과정중적작용。방법건립T2DM대서동물모형。채용쌍항체협심매련면역법검측혈청급심기APN수평,병채용면역조화적방법측정심기지련소수체1(AdipoR1)적단백표체。결과여대조조비교,T2DM조혈청화심기APN수평하강,심기AdipoR1단백적표체수평강저,차이균유통계학의의(P<0.05)。상관성분석현시,T2DM조대서혈청APN여혈청이도소、은태모형이도소저항지수정부상관(r=-0.613,P<0.05;r=-0.637,P<0.05),심기AdipoR1단백표체량여심기APN정정상관(r=0.89,P<0.05),여HOMA은태모형(HOMA-IR)급공복이도소수평(FINS)정부상관(r=-0.697,P<0.05;r=-0.593,P<0.05)。결론당뇨병심기병대서혈청급심기APN수평하강,심기AdipoR1단백표체하강,이도소저항가능재당뇨병심기병적발병과정중기도작용。
Objective To discuss the effects of expressions of adiponectin and adiponectin receptor 1 (AdipoR1) in the development of cardiomyopathy through observing them in rats with T2DM. Methods The model of T2DM was established in rats, and the level of serum and myocardial APN was detected by using double-antibody sandwich ELISA, and expression of AdipoR1 protein was detected by using immunohistochemistry technique. Results Compared with control group, the level of serum and myocardial APN decreased, and expression of AdipoR1 protein decreased in T2DM group (P<0.05). The correlation analysis showed that serum APN was negatively correlated to serum insulin and HOMA-insulin resistance (HOMA-IR, r=-0.613, P<0.05;r=-0.637, P<0.05). The expression of AdipoR1 protein was positively correlated to myocardial APN (r=0.89, P<0.05), and negatively correlated to HOMA-IR and fasting insulin (r=-0.697, P<0.05; r=-0.593, P<0.05). Conclusion The level of serum and myocardial APN, and expression of AdipoR1 protein decrease in rats with diabetic cardiomyopathy. IR may play a role in the development of diabetic cardiomyopathy.
