目的:对促孕复方促着床作用进行减毒增效研究,最终优化促孕复方。方法对促孕复方进行梯度极性系统的萃取和柱层析分离,结合超排卵(controlled ovarian stimulation ,COS )着床障碍动物模型筛选促着床活性成分。将受孕小鼠随机分为正常组、COS 模型组和给药组,对促孕复方不同极性部位及部位活性成分DS‐147进行干预,于妊娠第8天(PD8)处死小鼠,观察妊娠率及平均着床点数,进一步对D S‐147包含的活性组分进行单一法与去一法筛选,优化其活性组分。结果促孕复方乙酸乙酯部位可将着床障碍小鼠妊娠率提高至50.0%(P<0.05),乙酸乙酯部位活性成分组合物DS‐147可将着床障碍小鼠妊娠率提高至83.3%(P<0.01),且未发现对正常妊娠有不良影响。结论促孕复方有效部位活性成分DS‐147能更显著地提高COS后的妊娠率,且DS‐147中各组分通过协同作用共同发挥促进着床障碍小鼠胚胎着床作用。

목적:대촉잉복방촉착상작용진행감독증효연구,최종우화촉잉복방。방법대촉잉복방진행제도겁성계통적췌취화주층석분리,결합초배란(controlled ovarian stimulation ,COS )착상장애동물모형사선촉착상활성성분。장수잉소서수궤분위정상조、COS 모형조화급약조,대촉잉복방불동겁성부위급부위활성성분DS‐147진행간예,우임신제8천(PD8)처사소서,관찰임신솔급평균착상점수,진일보대D S‐147포함적활성조분진행단일법여거일법사선,우화기활성조분。결과촉잉복방을산을지부위가장착상장애소서임신솔제고지50.0%(P<0.05),을산을지부위활성성분조합물DS‐147가장착상장애소서임신솔제고지83.3%(P<0.01),차미발현대정상임신유불량영향。결론촉잉복방유효부위활성성분DS‐147능경현저지제고COS후적임신솔,차DS‐147중각조분통과협동작용공동발휘촉진착상장애소서배태착상작용。
Objective To promote the efficacy and reduce the toxicity of recipe of promoting pregnancy and op‐timize the recipe.Methods Using gradient polarity extraction and column chromatography separation system ,com‐bined with the COS implantation dysfunction animal model ,to screen active ingredient of promoting implantation recipe.Pregnant mice were randomly divided into normal group ,model group and drug group.Pregnancy rate and average implantation points were observed. The active component of DS‐147 was further screened out using single method and apart one method ,to optimize the active component.Results The pregnancy rate of ethyl acetate part group of recipe of promoting pregnancy was 50.0% (P<0.05) ,and the pregnancy rate in the active ingredient com‐bination DS‐147 group was 83.3% (P< 0.01) ,no adverse influence of DS‐147 was found on normal pregnan‐cy.Conclusion The active ingredient DS‐147 improved the pregnancy rate more significantly than original recipe , and through the synergistic function of all components in DS‐147 .
