目的:应用脓毒症患者外周血单个核细胞与LPS及IL-23共培养诱导干扰素-γ表达,探讨脓毒症单个核细胞免疫状态的改变。方法选取35例脓毒症患者作为脓毒症组,31例全身炎症反应综合征( SIRS)患者为SIRS组,17例健康对照者为对照组,取外周血浆分离单个核细胞( PBMCs),分别与LPS、IL-23及LPS+IL-23共同培养,72h后用ELISA法检测培养上清中IFN-γ的水平。结果 LPS、IL-23及LPS+IL-23与PBMCs细胞共同培养后,正常组可诱导PBMCs产生大量IFN-γ,脓毒症组和SIRS组与正常组相比IFN-γ的产生明显减少(p均<0.05)。结论脓毒症患者单个核细胞IFN-γ释放能力明显下降,表明脓毒症患者免疫状态受到抑制。
目的:應用膿毒癥患者外週血單箇覈細胞與LPS及IL-23共培養誘導榦擾素-γ錶達,探討膿毒癥單箇覈細胞免疫狀態的改變。方法選取35例膿毒癥患者作為膿毒癥組,31例全身炎癥反應綜閤徵( SIRS)患者為SIRS組,17例健康對照者為對照組,取外週血漿分離單箇覈細胞( PBMCs),分彆與LPS、IL-23及LPS+IL-23共同培養,72h後用ELISA法檢測培養上清中IFN-γ的水平。結果 LPS、IL-23及LPS+IL-23與PBMCs細胞共同培養後,正常組可誘導PBMCs產生大量IFN-γ,膿毒癥組和SIRS組與正常組相比IFN-γ的產生明顯減少(p均<0.05)。結論膿毒癥患者單箇覈細胞IFN-γ釋放能力明顯下降,錶明膿毒癥患者免疫狀態受到抑製。
목적:응용농독증환자외주혈단개핵세포여LPS급IL-23공배양유도간우소-γ표체,탐토농독증단개핵세포면역상태적개변。방법선취35례농독증환자작위농독증조,31례전신염증반응종합정( SIRS)환자위SIRS조,17례건강대조자위대조조,취외주혈장분리단개핵세포( PBMCs),분별여LPS、IL-23급LPS+IL-23공동배양,72h후용ELISA법검측배양상청중IFN-γ적수평。결과 LPS、IL-23급LPS+IL-23여PBMCs세포공동배양후,정상조가유도PBMCs산생대량IFN-γ,농독증조화SIRS조여정상조상비IFN-γ적산생명현감소(p균<0.05)。결론농독증환자단개핵세포IFN-γ석방능력명현하강,표명농독증환자면역상태수도억제。
Objective To investigate the changes of cellular immunity in patients with sepsis by co-culturing peripheral blood mononuclear cells and LPS and IL-23 were to induce the expression of IFN-γ. Methods The study selected 35 patients with sepsis as the sepsis group, 31 patients with systemic inflammatory response syndrome as the SIRS group and 17 healthy people as the controls. PBMCs were co-cultured with LPS, IL-23 and LPS+IL-23. 72 hours after, the level of IFN-γ was measured by ELISA. Results LPS, IL-23, LPS+IL-23 and PBMCs were co-cultured, which resulted in significant production of IFN-γby PBMC in the control group. Compared with the control group, the expression of IFN-γin the SIRS and sepsis groups was dramatically reduced(P<0. 05. Conclusion The expression of IFN-γ decreases, showing that the cellular immunity is inhibited in patients with sepsis.
