目的:对急性心肌梗死(AMI)患者白介素-10(IL-10)基因启动子-592C/A进行多态性分析,探讨AMI患者IL-10基因-592C/A单核苷酸多态性分布及其与AMI发病的相关性。方法采用DNA试剂盒提取170例AMI患者(AMI组)及153例排除冠心病患者(对照组)的外周静脉血DNA,经聚合酶链反应(PCR)扩增目的DNA片段后,用试剂盒回收PCR产物,测序鉴定IL-10基因启动子-592C/A多态性,并检测血糖和血脂等生化指标的变化。结果 IL-10基因启动子-592C/A在AMI组和对照组中均存在CC、AC和AA 3种基因型;两组患者3种基因型分布频率及A、C等位基因频率比较,差异均无统计学意义(P>0.05);经对多个危险因素的多元logistic回归分析显示,IL-10基因启动子-592C/A 3种基因型与AMI的发病不相关。结论 IL-10基因启动子-592C/A多态性可能与AMI的发病无明显关系。

Objective To explore analyze the polymorphism of interleukin-10(IL-10)gene-592C/A of patients with acute myocardial infarction(AMI) and its relevance to the occurrence of AMI by analyzing the polymorphism of IL-10 gene promoter-592C/A. Methods DNA kit was adopted to extract the DNA from peripheral venous blood of 170 AMI patients(AMI group) and 153 controls (control group) at the exclusion of coronary heart disease (CHD),after amplifying DNA segments by polymerases chain reaction(PCR),the kit was used to recycle reaction products of PCR to identify the polymorphism of IL-10 gene-529C/A and detect the biochemical indicators including blood glucose and blood lipid. Results IL-10 gene-592C/A had 3 genotypes(CC, AC and AA) in both AMI group and control group;there was no statistically significant difference on distribution frequencies of the 3 genotypes and allele frequencies of A and C between the AMI group and control group (P>0.05);the results of multivariate logis-tic regression analysis in multiple risk factors showed that the genotypes of IL-gene-592C/A were not revelent to the occurrence of AMI. Conclusion The polymorphism of IL-10 gene-592C/A has no relationship with AMI in the crowds of Changzhou region.
