Gynostemma pentaphyllum%CYP450%GST%UGT%rat
SD 大鼠自由饮用绞股蓝汁(绞股蓝汁每天新鲜配制,浓度为每100 g 水2 g 茶叶,100℃的水温浸泡30 min,取上清液),连续给药60 d,取出肝脏,用差速离心法制备肝脏胞浆液及肝脏微粒体,采用双光束紫外分光光度法测定 CYP3A、CYP2E1、NADPH-细胞色素 C 还原酶、UGT、GST 的活性及细胞色素 b5的含量,结果显示绞股蓝可显著升高细胞色素 b5的含量,显著诱导 CYP3A、UGT、GST、NADPH-细胞色素 C 还原酶的活性,但对 CYP2E1没有影响。提示绞股蓝与药物合用时,在体内可能会发生代谢性药物相互作用。

SD 대서자유음용교고람즙(교고람즙매천신선배제,농도위매100 g 수2 g 다협,100℃적수온침포30 min,취상청액),련속급약60 d,취출간장,용차속리심법제비간장포장액급간장미립체,채용쌍광속자외분광광도법측정 CYP3A、CYP2E1、NADPH-세포색소 C 환원매、UGT、GST 적활성급세포색소 b5적함량,결과현시교고람가현저승고세포색소 b5적함량,현저유도 CYP3A、UGT、GST、NADPH-세포색소 C 환원매적활성,단대 CYP2E1몰유영향。제시교고람여약물합용시,재체내가능회발생대사성약물상호작용。
The SD rats were maintained on freshly prepared aqueous extract of Gynostemma pentaphyllum (2%) while the control rats were given distilled water for 60 days.Rat liver was taken and liver microsome and cytochyma enzymes was obtained by different velocity centrifugation.Then the total contents and activities of CYP450 and GST, UGT of rats determined by double wavelength ultraviolet spectrophotometry.The results showed that the G .pen-taphyllum could increase the content of b5 ;Increase the activity of CYP3A,UGT,GST,NADPH-cytochtome C re-ductase;But had no effect on CYP2E1.The study showed that co-administrated with those drugs metabolized by the above major metabolizing enzymes in liver,metabolic herb-dtug interactions would happened.
