目的:探讨外源性甲胎蛋白( AFP)对体外培养的肝癌细胞系的生物学功能及意义。方法该实验选用了HepG2(AFP阳性)和HLE(AFP阴性)两种常见的肝癌细胞系,再加入不同浓度的外源性AFP处理24 h后,用MTT检测细胞的增殖情况;在这两种细胞系中加入凋亡诱导剂ATRA处理的同时加入外源性AFP,检测AFP对于细胞的抗凋亡能力的影响。结果在HepG2和HLE细胞中,加入不同浓度的外源性AFP后, MTT结果显示AFP可以明显促进肝癌细胞的增殖;而加入凋亡诱导剂ATRA之后,FCM结果显示细胞凋亡明显增加,加入外源性AFP后,细胞凋亡的情况明显好转,外源性AFP可以增强肝癌细胞的抗凋亡能力。结论外源性AFP不仅可以促进肝癌细胞系的增殖,同时还可以增强肝癌细胞的抗凋亡能力。

목적:탐토외원성갑태단백( AFP)대체외배양적간암세포계적생물학공능급의의。방법해실험선용료HepG2(AFP양성)화HLE(AFP음성)량충상견적간암세포계,재가입불동농도적외원성AFP처리24 h후,용MTT검측세포적증식정황;재저량충세포계중가입조망유도제ATRA처리적동시가입외원성AFP,검측AFP대우세포적항조망능력적영향。결과재HepG2화HLE세포중,가입불동농도적외원성AFP후, MTT결과현시AFP가이명현촉진간암세포적증식;이가입조망유도제ATRA지후,FCM결과현시세포조망명현증가,가입외원성AFP후,세포조망적정황명현호전,외원성AFP가이증강간암세포적항조망능력。결론외원성AFP불부가이촉진간암세포계적증식,동시환가이증강간암세포적항조망능력。
Objective To investigate the biological functions and significance of exogenous AFP in hepatoma cell lines.Methods Two types of normal hepatoma cell lines ( HepG2 and HLE) were chosen,added with different concentrations of exogenous AFP for 24 hours,MTT was used to detect cells proliferation;ATRA and AFP were employed to stimulate cells at the same time,FCM was used to detect cells apoptosis.Results For the HepG2 and HLE cells with exogenous AFP, MTT results showed that AFP could increase cell proliferation significantly.Comparing with control group,when cells were treated with ATRA alone,cells apoptosis were significantly increased,while after adding exogenous AFP,cell apoptosis were partially reversed.Conclusion Exogenous AFP can not only promote the proliferation of hepatoma cell lines,but also enhance the cells anti-apoptosis ability.
