目的:探索不同浓度的丙氨酸-谷氨酰胺二肽在序贯培养基及一程式培养基中的培养效果和最佳浓度。方法丙氨酸-谷氨酰胺二肽分别按0.7、0.85、1.0、1.15和1.3mmol/L配制序贯系列培养基的囊胚培养液和一程式培养基,将上述培养基分组分别用于小鼠受精卵的体外常规培养,100~104h后,观察其囊胚形成率和孵化率,并以商品化的GⅢ培养基作对照组。结果在序贯培养基中除1.3mmol/l组的囊胚形成率和孵化率显著降低以外,其余各组件均为显著性差异;在一程式培养基中,0.7、0.85和1.0mmol/L组的囊胚形成率明显高于其它各组,但0.7mmol/L组的孵出率显著高于其它各组。结论在体外培养基中,丙氨酸-谷氨酰胺二肽的浓度低于1.0mmol/L时可能更有利于体外培养中小鼠胚胎的发育。

목적:탐색불동농도적병안산-곡안선알이태재서관배양기급일정식배양기중적배양효과화최가농도。방법병안산-곡안선알이태분별안0.7、0.85、1.0、1.15화1.3mmol/L배제서관계렬배양기적낭배배양액화일정식배양기,장상술배양기분조분별용우소서수정란적체외상규배양,100~104h후,관찰기낭배형성솔화부화솔,병이상품화적GⅢ배양기작대조조。결과재서관배양기중제1.3mmol/l조적낭배형성솔화부화솔현저강저이외,기여각조건균위현저성차이;재일정식배양기중,0.7、0.85화1.0mmol/L조적낭배형성솔명현고우기타각조,단0.7mmol/L조적부출솔현저고우기타각조。결론재체외배양기중,병안산-곡안선알이태적농도저우1.0mmol/L시가능경유리우체외배양중소서배태적발육。
Objective To explore the effect of different concentrations of alanine-glutamine dipep-tide in sequential medium and program medium, and the best concentration in the medium. Methods Sequential medium series blastocyst culture medium and program medium with different final concentration of alanine-glutamine dipeptide in them: 0.7mmol/L、0.85mmol/L、1.0mmol/L、1.15mmol/L and 1.3mmol/L were prepared respectively. The above medium groups were used for mouse fertilized eggs conventional culture in vitro. The blastocyst formation rate and hatching rate was observed after 100~104 hours with the commercialization of G Ⅲ medium used as the con-trol group. Results Significant difference between groups were observed in the sequential medium group with the exception for final concentration of 1.3mmol/L, showing significantly decreased blastocyst formation rate and hatching rate; In the program medium, the blastocyst formation rate was obviously higher in 0.7mmol/L, 0.85mmol/L and 1.0mmol/L groups than other groups, and the hatching rate was significantly higher in 0.7mmol/L group than in other groups. Conclusion It might be more helpful in vitro culture of mouse embryos when alanine glutamine-dipeptide con-centrations below 1.0mmol/L in vitro culture medium.
