目的:观察不同昼夜节律下心力衰竭兔心肌细胞自发性钙火花发生率、时空特性及对交感刺激的反应。方法家兔心衰模型采用主动脉返流加主动脉缩窄的方法制作,依据相隔12 h的昼夜节律将心衰动物分为静息组及活动组,并分别按时间点分离心肌细胞(每组24个细胞),使用激光共聚焦扫描技术制作细胞悬浊液并进行观察,测量钙火花发生率、幅值、半高宽、半高时程、上升时间和半衰减时间的变化。结果静息组心肌细胞自发性钙火花发生率及钙火花幅值、半高宽、半高时程、上升时间和半衰减时间明显低于活动组(P<0.05);给予ISO灌流后,静息组心肌细胞钙火花发生率及时空特性参数明显高于活动组(P<0.05)。结论昼夜节律可影响衰竭心肌细胞局部钙释放及交感反应性,其可能是夜间或静息状态下高发室性心律失常的机制之一。
目的:觀察不同晝夜節律下心力衰竭兔心肌細胞自髮性鈣火花髮生率、時空特性及對交感刺激的反應。方法傢兔心衰模型採用主動脈返流加主動脈縮窄的方法製作,依據相隔12 h的晝夜節律將心衰動物分為靜息組及活動組,併分彆按時間點分離心肌細胞(每組24箇細胞),使用激光共聚焦掃描技術製作細胞懸濁液併進行觀察,測量鈣火花髮生率、幅值、半高寬、半高時程、上升時間和半衰減時間的變化。結果靜息組心肌細胞自髮性鈣火花髮生率及鈣火花幅值、半高寬、半高時程、上升時間和半衰減時間明顯低于活動組(P<0.05);給予ISO灌流後,靜息組心肌細胞鈣火花髮生率及時空特性參數明顯高于活動組(P<0.05)。結論晝夜節律可影響衰竭心肌細胞跼部鈣釋放及交感反應性,其可能是夜間或靜息狀態下高髮室性心律失常的機製之一。
목적:관찰불동주야절률하심력쇠갈토심기세포자발성개화화발생솔、시공특성급대교감자격적반응。방법가토심쇠모형채용주동맥반류가주동맥축착적방법제작,의거상격12 h적주야절률장심쇠동물분위정식조급활동조,병분별안시간점분리심기세포(매조24개세포),사용격광공취초소묘기술제작세포현탁액병진행관찰,측량개화화발생솔、폭치、반고관、반고시정、상승시간화반쇠감시간적변화。결과정식조심기세포자발성개화화발생솔급개화화폭치、반고관、반고시정、상승시간화반쇠감시간명현저우활동조(P<0.05);급여ISO관류후,정식조심기세포개화화발생솔급시공특성삼수명현고우활동조(P<0.05)。결론주야절률가영향쇠갈심기세포국부개석방급교감반응성,기가능시야간혹정식상태하고발실성심률실상적궤제지일。
Objective To observe the variation of the incidence and spatiotemporal properties of spontane-ous calcium spark in myocardial cells of rabbits with heart failure and their response toβ-Adrenergic stimulation with different circadian rhythms. Methods Rabbit models of heart failure were induced by combined aortic insufficiency and constriction, which were divided into resting group and active group according to circadian rhythms. Myocytes were isolated at 2 opposing time points (24 cells in each group). Cell suspension was prepared by confocal laser scan-ning and the observed. Incidence of calcium sparks and its spatiotemporal properties were evaluated by confocal scan-ning. Results Incidence rate, amplitude, full half-height width (FWDH), full duration at half maximum (FDHM), rising time (RT) and time to half-regression (THR) of spontaneous calcium spark in myocardial cells in resting group was markedly lower than that in active group (P<0.05). Underβ-Adrenergic stimulation, the incidence rate and properties of spontaneous calcium spark in myocardial cells in the resting group were significantly greater than that in the active group. Conclusion Circadian rhythms might influence the local calcium release and sympathetic reaction in failing heart cells, which could be a mechanism of high-incident ventricular arrhythmias at night or resting state.
