目的：研究唑来膦酸对胃癌细胞株SGC-7901的体外增殖抑制作用及对血管内皮生长因子(VEGF)的影响。方法采用CCK-8试验检测唑来膦酸对胃癌细胞株SGC-7901的生长抑制作用；酶联免疫吸附试验(ELISA)和半定量RT-PCR法检测药物对VEGF表达水平的变化。结果0.2~3.2μg/L的唑来膦酸能显著抑制SGC-7901细胞的增殖，且具有时间及剂量依赖性，不同药物浓度作用后，胃癌细胞株VEGF的蛋白及基因表达水平均明显降低(P<005)，具有浓度依赖性。结论唑来膦酸能抑制胃癌细胞株SGC-7901的生长增殖，其作用机制可能为下调VEGF表达。
목적：연구서래련산대위암세포주SGC-7901적체외증식억제작용급대혈관내피생장인자(VEGF)적영향。방법채용CCK-8시험검측서래련산대위암세포주SGC-7901적생장억제작용；매련면역흡부시험(ELISA)화반정량RT-PCR법검측약물대VEGF표체수평적변화。결과0.2~3.2μg/L적서래련산능현저억제SGC-7901세포적증식，차구유시간급제량의뢰성，불동약물농도작용후，위암세포주VEGF적단백급기인표체수평균명현강저(P<005)，구유농도의뢰성。결론서래련산능억제위암세포주SGC-7901적생장증식，기작용궤제가능위하조VEGF표체。
Objective To investigate the proliferation inhibition function of zoledronic acid on human gas-tric cancer cell SGC-7901 and its influence on vascular endothelial growth factor (VEGF). Methods CCK-8 cell vi-ability assay was performed to investigate the effect of zoledronic acid on cell proliferation. The expression of VEGF was examined by ELISA and semi-quantitative RT-PCR. Results Zoledronic acid inhibited the proliferation of SGC-7901 cells at a dose-and-time dependent manner (concentration range: 0.2~3.2μg/L). Meanwhile, ELISA and semi-quantitative RT-PCR results showed that the expression of VEGF was down-regulated at a dose-dependent fash-ion in SGC-7901 cells. Conclusion Zoledronic acid might inhibit the proliferation of SGC-7901 cells through down-regulation of VEGF.
