目的:观察微小RNA 100(miR-100)、微小RNA 148a(miR-148a)在肺癌细胞与组织中的表达,并探讨其临床意义。方法采用RT-PCR方法检测人肺癌细胞系NCI-H1299、NCI-H358、H460、A549及人正常支气管上皮细胞系HBE,以及肺癌及其癌旁组织中miR-100、miR-148a表达;分析肺癌组织中miR-100、miR-148a表达与其临床病理参数的关系。结果与HBE细胞相比,miR-100在NCI-H1299、NCI-H358及A549细胞中表达升高,在H460细胞表达降低,P均<0.05;miR-148a在NCI-H358、H460细胞中表达升高,在A549细胞中表达降低,P均<0.05。与癌旁组织相比,肺癌组织中miR-100表达降低,但差异无统计学意义(P=0.400);miR-148a表达升高(P=0.036)。肺癌组织中miR-100、miR-148a表达水平与患者性别、年龄、病理类型无关;有淋巴结转移者肺癌组织中miR-100表达水平较无淋巴结转移者低(P=0.016)。结论 miR-100、miR-148a在肺癌细胞中表达异常;在肺癌组织中,miR-100表达降低,miR-148a表达增加,二者可能参与肺癌的发生、发展。

목적:관찰미소RNA 100(miR-100)、미소RNA 148a(miR-148a)재폐암세포여조직중적표체,병탐토기림상의의。방법채용RT-PCR방법검측인폐암세포계NCI-H1299、NCI-H358、H460、A549급인정상지기관상피세포계HBE,이급폐암급기암방조직중miR-100、miR-148a표체;분석폐암조직중miR-100、miR-148a표체여기림상병리삼수적관계。결과여HBE세포상비,miR-100재NCI-H1299、NCI-H358급A549세포중표체승고,재H460세포표체강저,P균<0.05;miR-148a재NCI-H358、H460세포중표체승고,재A549세포중표체강저,P균<0.05。여암방조직상비,폐암조직중miR-100표체강저,단차이무통계학의의(P=0.400);miR-148a표체승고(P=0.036)。폐암조직중miR-100、miR-148a표체수평여환자성별、년령、병리류형무관;유림파결전이자폐암조직중miR-100표체수평교무림파결전이자저(P=0.016)。결론 miR-100、miR-148a재폐암세포중표체이상;재폐암조직중,miR-100표체강저,miR-148a표체증가,이자가능삼여폐암적발생、발전。
Objective To investigate the expression and clinical significance of miR-100 and miR-148a in lung cancer cells and tissues.Methods RT-PCR method was used to detect the expression of miR-100 and miR-148a in lung cancer cell lines NCI-H1299, NCI-H358, H460, A549 and normal human bronchial epithelial cell line HBE, and in the lung cancer tissues and adjacent tissues.To analyze the relationship between the expression of miR-100 and miR-148a and clini-copathological parameters of lung cancer.Results Compared with HBE cells, miR-100 expression in NCI-H1299, NCI-H358 and A549 increased significantly, decreased in H460, all P<0.05; the expression of miR-148a in NCI-H358 and H460 increased, decreased in A549, all P<0.05.Compared to non-tumor tissues, miR-100 expression was reduced in lung cancer, but the difference was not statistically significant (P=0.400);while the expression of miR-148a was signifi-cantly increased (P=0.036).miR-100 expression in lung cancer tissue with lymph node metastasis was lower than that in lung cancer tissue without lymph node metastasis (P=0.016).Conclusions The expression of miR-148a, miR-100 were aberrantly changed in lung cancer cells.miR-100 expression decreased in lung cancer tissues, miR-148a increased, which both maight be involved in the tumorigenesis of lung cancer.
