目的:探讨壮医药线点灸联合针挑疗法对大鼠坐骨神经痛相关神经元和递质的影响。方法将 SPF 级 SD 大鼠80只随机分为正常对照组(简称对照组)、假手术组、模型组、药线+针挑组(简称联合组)共4组,每组20只。模型组建立坐骨神经慢性压迫损伤模型,联合组在模型组造模基础上,采用药线点灸、针挑联合应用的干预措施,连续干预21 d。然后检测活性氧(ROS)、超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH -Px)活性或水平,检测神经递质γ-氨基丁酸(Gamma-aminobutyric acid,GABA)和5-羟色胺(5- HT)含量、GABA A 受体(GABAA R) mRNA 表达以及观察 GABA 能神经元的凋亡变化。结果与对照组比较,模型组血清 ROS、MDA 水平均显著上升,差异有统计学意义(P <0.01),GSH-Px、SOD 活性和 GABA、5- HT 含量均显著下降,差异有统计学意义(P <0.01), GABAA R mRNA 表达下降,差异有统计学意义(P <0.05),凋亡细胞较正常组多。与模型组比较,联合组 ROS、MDA水平均显著下降,差异有统计学意义(P <0.01),GSH-Px、SOD 活性和 GABA、5- HT 含量上升或显著上升,差异有统计学意义(P <0.05或 P <0.01),GABAA R mRNA 表达上升,差异有统计学意义(P <0.05),凋亡细胞较模型组下降。结论壮医药线点灸联合针挑疗法保护 GABA 神经元及其功能,可能与该疗法能增强抗氧化酶活性,防止氧化损伤 GABA 能神经元有关。

목적:탐토장의약선점구연합침도요법대대서좌골신경통상관신경원화체질적영향。방법장 SPF 급 SD 대서80지수궤분위정상대조조(간칭대조조)、가수술조、모형조、약선+침도조(간칭연합조)공4조,매조20지。모형조건립좌골신경만성압박손상모형,연합조재모형조조모기출상,채용약선점구、침도연합응용적간예조시,련속간예21 d。연후검측활성양(ROS)、초양화물기화매(SOD)、병이철(MDA)、곡광감태과양화물매(GSH -Px)활성혹수평,검측신경체질γ-안기정산(Gamma-aminobutyric acid,GABA)화5-간색알(5- HT)함량、GABA A 수체(GABAA R) mRNA 표체이급관찰 GABA 능신경원적조망변화。결과여대조조비교,모형조혈청 ROS、MDA 수평균현저상승,차이유통계학의의(P <0.01),GSH-Px、SOD 활성화 GABA、5- HT 함량균현저하강,차이유통계학의의(P <0.01), GABAA R mRNA 표체하강,차이유통계학의의(P <0.05),조망세포교정상조다。여모형조비교,연합조 ROS、MDA수평균현저하강,차이유통계학의의(P <0.01),GSH-Px、SOD 활성화 GABA、5- HT 함량상승혹현저상승,차이유통계학의의(P <0.05혹 P <0.01),GABAA R mRNA 표체상승,차이유통계학의의(P <0.05),조망세포교모형조하강。결론장의약선점구연합침도요법보호 GABA 신경원급기공능,가능여해요법능증강항양화매활성,방지양화손상 GABA 능신경원유관。
Objective To investigate the effects of Zhuang medical thread moxibustion combined with Zhuang acupuncture on neurons and neurotransmitters in rats with sciatica. Methods Eighty SD rats with SPF grade were randomly assigned to one of the four groups:a control group (n =20),a sham group(n =20),a model group(n =20),and a medical thread moxibustion combined acupuncture group(combined group, n =20).Models of chronic constriction injury of sciatic nerve were set up in the model group,and based on the models,the combined group was treated with Zhuang medical thread moxibustion combined with Zhuang acu-puncture for twenty-one days continuously.Then the levels of ROS,MDA and activities of SOD,GSH-Px were measured and the contents of GABA and 5-HT,the expression of GABAAR mRNA were examined,the apoptosis changes of GABAergic neurons were observed. Results Compared to the control group,in the model group the levels of ROS,MDA increased significantly (P <0.01),activities of SOD,GSH-Px and the contents of GABA and 5 - HT all reduced significantly (P <0.01),the expression of GABAAR mRNA re-duced (P <0.05),more apoptotic cells exhibited .Compared to the model group,in the combined group the levels of ROS,MDA reduced significantly (P <0.01),activities of SOD,GSH-Px and the contents of GA-BA and 5-HT increased (P <0.05 or P <0.01),the expression of GABAAR mRNA increased (P <0.05),apoptotic cells reduced. Conclusion The treatment of Zhuang medical thread moxibustion combined with Zhuang acupuncture could protect GABAergic neurons and its function,which might be associated with enhan-cing activities of antioxidant enzyme for preventing oxidative damages in GABAergic neurons.
