目的:探讨趋化素样因子1(CKLF1)拮抗肽C19局部皮下注射对特应性皮炎小鼠模型的作用。方法用卵清蛋白( OVA)经皮致敏BALB/c小鼠建立特应性皮炎模型。将60只BALB/c小鼠随机分成特应性皮炎组、对照组、预防组和治疗组,每组15只。观察小鼠致敏局部皮肤及组织病理改变。以ULSTM荧光标定技术检测各组总RNA中mi-cro RNA的表现图谱。结果①与对照组及预防组相比,皮炎组小鼠局部皮肤出现红斑、鳞屑等皮损,治疗组小鼠皮损经治疗后红斑、水肿减轻,鳞屑减少。小鼠局部皮损组织病理学显示,经CKLF1拮抗肽C19预防或治疗后,小鼠表皮增厚、细胞间水肿、真皮血管扩张充血及炎性细胞浸润均较轻。②以ULSTM荧光标定技术检测各组总RNA中micro RNA的表现图谱,通过鉴定各组间上调与下调的特定 micro RNA 基因,筛选调节炎症细胞因子特定靶基因表达的 micro RNA,初步分析发现,经CKLF1拮抗肽C19局部外用治疗或预防后,小鼠皮损中部分调节炎症细胞因子特定基因表达的micro RNA上调。结论 CKLF1拮抗肽C19可能在预防或治疗特应性皮炎中具有重要作用,为了解及探索治疗特应性皮炎的多肽类药物提供了新途径。
目的:探討趨化素樣因子1(CKLF1)拮抗肽C19跼部皮下註射對特應性皮炎小鼠模型的作用。方法用卵清蛋白( OVA)經皮緻敏BALB/c小鼠建立特應性皮炎模型。將60隻BALB/c小鼠隨機分成特應性皮炎組、對照組、預防組和治療組,每組15隻。觀察小鼠緻敏跼部皮膚及組織病理改變。以ULSTM熒光標定技術檢測各組總RNA中mi-cro RNA的錶現圖譜。結果①與對照組及預防組相比,皮炎組小鼠跼部皮膚齣現紅斑、鱗屑等皮損,治療組小鼠皮損經治療後紅斑、水腫減輕,鱗屑減少。小鼠跼部皮損組織病理學顯示,經CKLF1拮抗肽C19預防或治療後,小鼠錶皮增厚、細胞間水腫、真皮血管擴張充血及炎性細胞浸潤均較輕。②以ULSTM熒光標定技術檢測各組總RNA中micro RNA的錶現圖譜,通過鑒定各組間上調與下調的特定 micro RNA 基因,篩選調節炎癥細胞因子特定靶基因錶達的 micro RNA,初步分析髮現,經CKLF1拮抗肽C19跼部外用治療或預防後,小鼠皮損中部分調節炎癥細胞因子特定基因錶達的micro RNA上調。結論 CKLF1拮抗肽C19可能在預防或治療特應性皮炎中具有重要作用,為瞭解及探索治療特應性皮炎的多肽類藥物提供瞭新途徑。
목적:탐토추화소양인자1(CKLF1)길항태C19국부피하주사대특응성피염소서모형적작용。방법용란청단백( OVA)경피치민BALB/c소서건립특응성피염모형。장60지BALB/c소서수궤분성특응성피염조、대조조、예방조화치료조,매조15지。관찰소서치민국부피부급조직병리개변。이ULSTM형광표정기술검측각조총RNA중mi-cro RNA적표현도보。결과①여대조조급예방조상비,피염조소서국부피부출현홍반、린설등피손,치료조소서피손경치료후홍반、수종감경,린설감소。소서국부피손조직병이학현시,경CKLF1길항태C19예방혹치료후,소서표피증후、세포간수종、진피혈관확장충혈급염성세포침윤균교경。②이ULSTM형광표정기술검측각조총RNA중micro RNA적표현도보,통과감정각조간상조여하조적특정 micro RNA 기인,사선조절염증세포인자특정파기인표체적 micro RNA,초보분석발현,경CKLF1길항태C19국부외용치료혹예방후,소서피손중부분조절염증세포인자특정기인표체적micro RNA상조。결론 CKLF1길항태C19가능재예방혹치료특응성피염중구유중요작용,위료해급탐색치료특응성피염적다태류약물제공료신도경。
Objective To explore the function of chemokine-like factor 1(CKLF1)antagonism peptides C19 in mouse model with atop-ic dermatitis. Methods The mouse model of atopic dermatitis were established by sensitization with ovalbumin( OVA)through the skin. 60 BALB/c mice were randomly divided into atopic dermatitis( AD)group,control group,prevention group,and treatment group,15 mice in each group. The gene expression profile of micro RNA in total RNA of every groups by ULSTM fluorescence detection calibration was detected. Results①The AD group showed local inflammation in the skin compared with the control group and prevention group. Epidermal thickening and signifi-cant inflammatory cellular infiltration were observed and the skin lesion of treatment group mice improved after 2 times treatment with C19. ② In order to detect the gene expression profile of micro RNA in total RNA of every groups by ULSTM fluorescence detection calibration,the data indi-cates that the expression level of micro RNA which regulate the specific gene of inflammatory cytokines in the lesions. The gene was up-regulated after treatment or prevention with C19. Conclusion Prevention or treatment with the CKLF1 antagonism peptides C19 may play an important ther-apeutic role in atopic dermatitis. Further study about the peptide may contribute to better understanding and exploring new anti-inflammatory medicine for the treatment of atopic dermatitis.
