目的:探讨中药红曲对高脂血症大鼠主动脉过氧化物酶体增殖物活化受体γ(PPARγ)和脂联素受体1和2(Adi-poR1/AdipoR2)mRNA的影响。方法 SPF级雄性SD大鼠36只,随机数字表法分为正常组(12只)和高脂组(24只),高脂饲料喂养12周后高脂组造模成功,随机分为模型组和红曲组,每组12只,分别予0.9%氯化钠溶液和红曲水提液灌胃4周,检测血脂水平, HE染色观察主动脉病理改变,Western boltting检测PPARγ表达,ELISA检测血清脂联素水平,实时荧光定量PCR检测 Adi-poR1/AdipoR2 mRNA的表达。结果高脂组TG、TC和LDL- C升高,HDL- C下降(P<0.05);给药4周后,与模型组比较,红曲组TG、TC和LDL- C降低(P<0.01或0.05),HDL- C升高(P<0.05);红曲组血清脂联素、AdipoR1和AdipoR2 mRNA的表达较模型组升高(P<0.05或0.01)。结论红曲能降低高脂血症大鼠血脂水平,其机制可能与增加主动脉PPARγ蛋白的表达,升高血清脂联素水平及AdipoR1和AdipoR2 mRNA表达有关。

목적:탐토중약홍곡대고지혈증대서주동맥과양화물매체증식물활화수체γ(PPARγ)화지련소수체1화2(Adi-poR1/AdipoR2)mRNA적영향。방법 SPF급웅성SD대서36지,수궤수자표법분위정상조(12지)화고지조(24지),고지사료위양12주후고지조조모성공,수궤분위모형조화홍곡조,매조12지,분별여0.9%록화납용액화홍곡수제액관위4주,검측혈지수평, HE염색관찰주동맥병리개변,Western boltting검측PPARγ표체,ELISA검측혈청지련소수평,실시형광정량PCR검측 Adi-poR1/AdipoR2 mRNA적표체。결과고지조TG、TC화LDL- C승고,HDL- C하강(P<0.05);급약4주후,여모형조비교,홍곡조TG、TC화LDL- C강저(P<0.01혹0.05),HDL- C승고(P<0.05);홍곡조혈청지련소、AdipoR1화AdipoR2 mRNA적표체교모형조승고(P<0.05혹0.01)。결론홍곡능강저고지혈증대서혈지수평,기궤제가능여증가주동맥PPARγ단백적표체,승고혈청지련소수평급AdipoR1화AdipoR2 mRNA표체유관。
Objective To investigate the effect of red kojic rice on expression of aorta peroxidase proliferator- activated receptorγ(PPARγ) and adiponectin receptor 1 and adiponectin receptor 2 (AdipoR1 and AdipoR2) mRNA in hyperlipidemic rat. Methods Thirty- six SPF male SD rats were randomly divided into control group (n=12) and hyperlipidemia group (n=24). Hyper-lipidemia was induced by feeding high- fat diet for 12 weeks. The hyperlipidemic rats were further divided into model group and treatment group (n=12 in each group), rats in two groups were given 0.9%normal saline and red kojic rice by gavage for 4 weeks, respectively. Serum lipid levels were measured and pathological changes in aorta were observed with HE staining. The expression of PPARγwas detected by Western blotting;serum adiponectin was measured by ELISA;AdipoR1 and AdipoR2 mRNA was de-tected by real time RT- PCR. Results After twelve weeks of high fat feeding, the triglyceride (TG), total cholesterol (TC) and low density lipoprotein cholesterol (LDL- C) in hyperlipidemia group increased significantly (P<0.01), and high density lipoprotein cholesterol (HDL- C) decreased (P<0.05). Four weeks after administration, compared with the model group, the TG, TC and LDL- C in the treatment group were significantly decreased (P<0.01 or 0.05), while HDL- C increased (P<0.05). The aortic intimal thickening and the proliferation of smooth muscle cel s were decreased, the expression of PPARγand AdipoR1/AdipoR2 mRNA was increased in treatment group compared to model group. Conclusion Red kojic rice can lower the levels of blood lipids in hy-perlipidemic rats, which may be related to u aortic PPARγprotein expression and AdipoR1/AdipoR2 mRNA expression.
