目的:检测同源盒基因B7( HOXB7)在肺腺癌中的表达情况,并探讨其临床意义。方法分别应用逆转录PCR( RT?PCR)和Western blotting法检测人正常肺支气管上皮细胞系HBE及4株肺腺癌细胞系A549、H1975、H1650、H322中HOXB7的表达情况。应用免疫组织化学染色及Western blotting法检测HOXB7在75例肺腺癌及配对癌旁正常组织中的表达,并分析其表达与肺腺癌临床病理特征及预后的关系。结果4株肺腺癌细胞系中HOXB7 mRNA和蛋白的表达水平均显著高于正常肺支气管上皮细胞系(P<0?05)。 HOXB7蛋白在肺腺癌及对应癌旁正常组织中的高表达率分别为76?0%(57/75)和9?3%(7/75),差异有统计学意义( P<0?05)。 HOXB7蛋白在肺腺癌组织中的表达水平与临床分期、病理分级及淋巴结转移有关( P<0?05),而与性别、年龄无关( P>0?05)。 HOXB7低表达者的中位生存期为68个月,高于高表达者的51个月,差异有统计学意义( P<0?05)。结论 HOXB7在肺腺癌中高表达,且与肿瘤的侵袭转移和疾病进展相关,有可能作为肺腺癌早期诊断及评估预后的新靶标。

Objective To detect the expression of homeobox genes( HOX) B7 in human lung adenocarcinoma, and investigate its clinical significance. Methods The expressions of HOXB7 mRNA and proteins in human lung adenocarcinoma cell lines A549, H1975, H1650, H322 and human bronchial epithelial cell line HBE were detected by reverse transcription PCR( RT?PCR) and West?ern blotting method, respectively. Immunohistochemistry staining was used to examine the expressions of HOXB7 in 75 samples of lung adenocarcinoma and their corresponding adjacent normal lung tissues. The correlation between HOXB7 expression and clinicopathological features and prognosis of lung adenocarcinoma were analyzed. Results The expression levels of HOXB7 mRNA and proteins in four lung adenocarcinoma cell lines were significantly higher than those in human bronchial epithelial cell line. The over?ex?pression rate of HOXB7 in lung adenocarcinoma and corresponding paracancerous tissue was 76?0%( 57/75) and 9?3%( 7/75) with statistical difference( P<0?05) . Statistical analysis revealed that HOXB7 protein expression was correlated with clinical stage, patholog?ical grade and lymph node metastasis in lung adenocarcinoma( P<0?05) . The median overall survival time of patients with low HOXB7 expression was 68 months, higher than 51 months of patients with over?expression with statistically significant( P<0?05) . Conclusion The over?expression of HOXB7 relates to the invasion, metastasis and progression of lung adenocarcinoma. It may play an important role in earlier diagnosis and prognosis evaluation for lung adenocarcinoma.
