目的：观察法舒地尔对急性有机磷中毒（acute organophosphorus pesticides poisoning，AOPP）大鼠横纹肌的保护作用。方法：清洁级雄性 Sprague Dawley 大鼠，采用经腹腔注射敌敌畏（dichlorvos，DDVP）法制备 AOPP 大鼠模型。将24只大鼠随机分成对照组、中毒组、干预组，每组各8只。麻醉后，行气管插管术，接动物人工呼吸机辅助通气。对照组经腹腔注射0．4 mL 0．9％氯化钠液；中毒组和干预组经腹腔注射30 mg／kg DDVP；干预组在麻醉前30 min 经腹腔注射法舒地尔10 mg／kg。每组大鼠在造模后2 h 采集血标本，应用自动生化分析仪检测血清心肌肌钙蛋白 T（cardiac troponin T，cTnT）水平；采用酶联免疫吸附试验（enzyme-linked immunosorbent assay，ELISA）法检测肌酸激酶同工酶 MM（creatine kinase isoenzyme MM，CK-MM）、乳酸脱氢酶同工酶 M （lactate dehydrogenase isoenzyme M，LDH-M）水平。采用逆转录-聚合酶链反应（reverse transcriptase polymerase chain reaction，RT-PCR）法检测左心室心肌、左侧腓肠肌 Rho 相关的卷曲螺旋型蛋白激酶 I（Rho-associated coiled-coil containing protein kinase I，ROCKI）mRNA 的表达。结果：与中毒组比较，干预组大鼠的横纹肌中毒症状减轻。与对照组比较，中毒组与干预组大鼠血清 cTnT 值均升高（P ＜0．05）。与中毒组比较，干预组血清 cTnT 值上升幅度下降（P ＜0．05）。对照组、中毒组与干预组大鼠血清 CK-MM、LDH-M 值差异均无统计学意义（P ＞0．05）。与对照组比较，中毒组大鼠左心室心肌与左侧腓肠肌 ROCKI mRNA 表达水平上升（P ＜0．05）；与中毒组比较，干预组大鼠左心室心肌与左侧腓肠肌 ROCKI mRNA 表达显著降低（P ＜0．05）。结论：法舒地尔通过抑制 ROCKI mRNA 的表达，减轻 AOPP 大鼠的横纹肌中毒症状，有保护横纹肌的作用。
목적：관찰법서지이대급성유궤린중독（acute organophosphorus pesticides poisoning，AOPP）대서횡문기적보호작용。방법：청길급웅성 Sprague Dawley 대서，채용경복강주사활활외（dichlorvos，DDVP）법제비 AOPP 대서모형。장24지대서수궤분성대조조、중독조、간예조，매조각8지。마취후，행기관삽관술，접동물인공호흡궤보조통기。대조조경복강주사0．4 mL 0．9％록화납액；중독조화간예조경복강주사30 mg／kg DDVP；간예조재마취전30 min 경복강주사법서지이10 mg／kg。매조대서재조모후2 h 채집혈표본，응용자동생화분석의검측혈청심기기개단백 T（cardiac troponin T，cTnT）수평；채용매련면역흡부시험（enzyme-linked immunosorbent assay，ELISA）법검측기산격매동공매 MM（creatine kinase isoenzyme MM，CK-MM）、유산탈경매동공매 M （lactate dehydrogenase isoenzyme M，LDH-M）수평。채용역전록-취합매련반응（reverse transcriptase polymerase chain reaction，RT-PCR）법검측좌심실심기、좌측비장기 Rho 상관적권곡라선형단백격매 I（Rho-associated coiled-coil containing protein kinase I，ROCKI）mRNA 적표체。결과：여중독조비교，간예조대서적횡문기중독증상감경。여대조조비교，중독조여간예조대서혈청 cTnT 치균승고（P ＜0．05）。여중독조비교，간예조혈청 cTnT 치상승폭도하강（P ＜0．05）。대조조、중독조여간예조대서혈청 CK-MM、LDH-M 치차이균무통계학의의（P ＞0．05）。여대조조비교，중독조대서좌심실심기여좌측비장기 ROCKI mRNA 표체수평상승（P ＜0．05）；여중독조비교，간예조대서좌심실심기여좌측비장기 ROCKI mRNA 표체현저강저（P ＜0．05）。결론：법서지이통과억제 ROCKI mRNA 적표체，감경 AOPP 대서적횡문기중독증상，유보호횡문기적작용。
Objective:To investigate the protective effect of fasudil on the striated muscle in rats with acute organophosphorus pesticides poisoning(AOPP).Methods:The AOPP model was established by peritoneal injection of dichlorvos (DDVP).Rats were randomly divided into control group(n=8),poisoned group(n=8)and pretreated group(n=8).All rats in the 3 groups were supported with breathing machine connected with the tube from incision of trachea.The control group was peritoneally in-jected 0.9% sodium chloride solution (0.4 mL).The poisoned group and the pretreated group were peritoneally injected DDVP (30 mg/kg).The pretreated group was peritoneally injected fasudil(10 mg/kg)30 min before anesthesia.Serum level of cardiac troponin T(cTnT)was measured by using autonomic biochemical analyzer,and creatine kinase isoenzyme MM(CK-MM)and lactate dehydrogenase isoenzyme M(LDH-M)were measured by enzyme-linked immunosorbent assay(ELISA)2 h later.The expression levels of Rho-associated coiled-coil containing protein kinase I (ROCKI)mRNA of left ventricular myocardium and left gastrocnemius muscle were analyzed by reverse transcriptase polymerase chain reaction(RT-PCR).Results:Compared with the poisoning group,the striated muscle poisoning symptoms were less serious in the pretreated group.Compared with the con-trol group,the level of cTnT in the poisoning group and the pretreated group significantly increased(P <0.05).Compared with the poisoning group,the level of cTnT in the pretreated group significantly decreased(P <0.05).The levels of CK-MM and LDH-M were not obviously different among 3 groups.Compared with the control group,the expression level of ROCKI mRNA of left ventricular myocardium and left gastrocnemius muscle in the poisoning group significantly increased(P <0.05).Com-pared with the poisoning group,the expression level of ROCKI mRNA of left ventricular myocardium and left gastrocnemius muscle in the pretreated group was significantly inhibited(P <0.05).Conclusions:Fasudil can relieve symptoms of AOPP in rats.Fasudil has protective effect on the striated muscle in rats.
