目的:观察Krüppel样转录因子KLF2在内毒素诱导大鼠急性肺损伤(ALI)时的动态表达,分析两者与急性肺损伤的相关性。方法将100只SD大鼠随机分为正常对照组和模型组,模型组进一步随机分成三组,模型组采用尾静脉注射脂多糖(LPS,5mg/kg)复制ALI模型,分别于尾静脉注射2h、4h、24h后采血及肺组织。观察各组病理表现、RT-PCR检测KLF2 mRNA在血清中及肺组织的表达,分析KLF2在急性肺损伤中的动态表达,采用SPSS17.0软件进行统计学分析。结果病理检查显示造模后4h肺损伤最为明显,KLF2 mRNA在正常大鼠肺组织及血清中均有明显表达,模型组各组KLF2的表达较对照组显著减弱(P<0.01),且KLF2 mRNA在2h表达明显低于4h组和24h组(P<0.01)。结论 KLF2在大鼠急性肺损伤表达变化早于病理变化,KLF2可作为急性肺损伤早期诊断的分子标志物。

목적:관찰Krüppel양전록인자KLF2재내독소유도대서급성폐손상(ALI)시적동태표체,분석량자여급성폐손상적상관성。방법장100지SD대서수궤분위정상대조조화모형조,모형조진일보수궤분성삼조,모형조채용미정맥주사지다당(LPS,5mg/kg)복제ALI모형,분별우미정맥주사2h、4h、24h후채혈급폐조직。관찰각조병리표현、RT-PCR검측KLF2 mRNA재혈청중급폐조직적표체,분석KLF2재급성폐손상중적동태표체,채용SPSS17.0연건진행통계학분석。결과병리검사현시조모후4h폐손상최위명현,KLF2 mRNA재정상대서폐조직급혈청중균유명현표체,모형조각조KLF2적표체교대조조현저감약(P<0.01),차KLF2 mRNA재2h표체명현저우4h조화24h조(P<0.01)。결론 KLF2재대서급성폐손상표체변화조우병리변화,KLF2가작위급성폐손상조기진단적분자표지물。
Objective To research the dynamic expression of KLF2 in the acute lung injury(ALI)rats induced by lipopolysaccharide(LPS),and analysis the correlation of KLF2 in ALI . Methods A total of 100 SD rats were randomly divided into 2 groups of normal control group and LPS treated group,then the latter group were randomly divided into 3 groups,which respectively taken serum and lung tissue samples 2h,4h and 24h after molding .The ALI model was induced by injecting 5mg/kg LPS in tail vein .To observe pathological manifestations of each group,detect KLF2mRNA expression in serum and lung tissue by RT-PCR,and analysis of dynamic expression,While using SPSS17.0 software for statistical analysis . Results The pathology shows most obvious damage of lung tissue is 4 hours after modeling,the expression of KLF2 mRNA in the lung tissue and serum of control group were clear,and significantly reduced in LPS treated group compared to control group(P<0.01),the expression of KLF2 in LPS treated group of 2hours was lower than groups of 4 hours and 24 hours(P<0.01).Conclusion The expression changes of KLF2 mRNA was earlier than the pathological changes in acute lung injury,which can be used as a predictive diagnostics candidate molecular markers of ALI.
