目的:观察阻断Notch通路对于大鼠骨髓间充质干细胞( Bone mesenchymal stem cells;BMSC )增殖及成骨分化的影响。方法:建立绝经后骨质疏松大鼠模型,对BMSC 进行分离鉴定及培养。设置阴性对照组及DAPT应用组,利用MTT法及ALP(Alkaline phosphatase)检测观察骨质疏松大鼠BMSC的增殖与成骨分化。结果:对照组及DAPT组BMSC 到第9d增殖达到最高峰。自第3d开始,DAPT组OD值要低于对照组(n=6,P<0.05)。对照组与DAPT组ALP 表达均在第7d达到峰值,第7d前两组之间ALP表达无明显差异(P>0.05),DAPT组在第7-11d时ALP 表达要高于对照组,具有统计学差异( n=6,P<0.01)。结论:DAPT阻断Notch通路后,骨质疏松大鼠BMSC的增殖受到了抑制,但对于诱导骨质疏松大鼠BMSC向成骨细胞定向分化的能力是增强的。

목적:관찰조단Notch통로대우대서골수간충질간세포( Bone mesenchymal stem cells;BMSC )증식급성골분화적영향。방법:건립절경후골질소송대서모형,대BMSC 진행분리감정급배양。설치음성대조조급DAPT응용조,이용MTT법급ALP(Alkaline phosphatase)검측관찰골질소송대서BMSC적증식여성골분화。결과:대조조급DAPT조BMSC 도제9d증식체도최고봉。자제3d개시,DAPT조OD치요저우대조조(n=6,P<0.05)。대조조여DAPT조ALP 표체균재제7d체도봉치,제7d전량조지간ALP표체무명현차이(P>0.05),DAPT조재제7-11d시ALP 표체요고우대조조,구유통계학차이( n=6,P<0.01)。결론:DAPT조단Notch통로후,골질소송대서BMSC적증식수도료억제,단대우유도골질소송대서BMSC향성골세포정향분화적능력시증강적。
Objective:To observe the effect of blocking notch signaling pathway on bone marrow mesen-chymal stem cell ( BMSC) proliferation and differentiation .Method:Established postmenopausal osteoporosis rat model first , and then BMSC were isolated for identification and cultivation .The rats were divided into shame group and DAPT group .observed BMSC proliferation and osteogenic differentiation of with MTT assay and the marker of ALP .Result:BMSC proliferation of both groups reached a peak value at the 9th day.OD values of DAPT group were lower than that of the control group from the 3rd day(n=6,P<0.05).The ALP expression of both groups reached a peak value at the 7th day.And at the 7th and 11th day, the ALP expres-sion of DAPT group were higher than control group (n=6,P <0.01).Conclusion:After blocking the notch pathway with DAPT, the BMSC proliferation of osteoporosis rat was inhibited , but the osteoblasts differentia-tion of BMSC was enhanced .
