目的:研究神经病理性疼痛( NP )对大鼠空间识别记忆功能与海马区组蛋白去乙酰化酶2( HADC2)表达的影响。方法将30只健康的Wistar雄性大鼠随机分为3组(每组10只),分别为对照组( CON组)、假手术组( SO组)、NP模型组( NP组)。 NP模型采用右侧坐骨神经慢性压迫( CCI)的方法制备。各组大鼠分别于术后3d,7d,10d和14d测右侧后爪机械缩足阈值( MWT)和热缩足潜伏期( TWL)。术后第14天,进行八臂迷宫实验观测NP对大鼠的空间识别记忆功能的影响。八臂迷宫实验完成后立即处死大鼠,通过Western Blot-ting方法测定海马区HADC2蛋白表达水平。结果与CON组比较,SO组和NP组的术后痛阈明显降低,空间识别记忆功能减退,HDAC2表达明显升高( P<0.05)。与SO组比较,NP组的术后痛阈降低,空间识别记忆功能减退,HADC2表达明显升高( P<0.05)。结论 NP可导致大鼠空间识别功能减退,同时海马区HDAC2表达升高。

[ ABSTRACT] Objective To evaluate the effect of neuropathy pain on spatial memory function of rats and the expression of histone deacetylase 2 in hippocampus.Methods The 30 healthy male Wister rats were randomly divided into 3 groups( n=10 in each group):con-trol group,sham operated group and neuropathic pain group.The NP models were prepared by the chronic constriction injury of the right sciat-ic nerve.Mechanical withdrawal threshold and thermal withdrawal latency of right hind legs were measured on 3th,7th,10th and 14th day af-ter operation respectively.On the 14th day after operation,eight-arm radial maze task was carried out to investigate the effect of neuropathy pain on spatial memory function in rats.Once the test was finished,rats were killed to measure the expression of HDAC2 protein in hippocam-pus by Western Blotting methods.Results Compared with CON group,the pain threshold was reduced significantly and spatial memory func-tion was significantly decreased in SO and NP group(P<0.05),while the HDAC2 expression level was significantly increased(P<0.05). Compared with SO group,the pain threshold was obviously reduced and the spatial memory function was significantly decreased in NP group (P<0.05),while the HDAC2 expression level was significantly increased(P<0.05).Conclusion The neuropathy pain can cause the de-crease of spatial memory function of rats and up-regulate the expression level of HDAC2 in hippocampus.
