以本课题组选育的10个审(鉴)定品种为材料,选用福建5个具有代表性品种为参照,利用 SSR 分子标记分析其遗传差异并建立分子指纹图谱,结果表明,从105对 SSR 引物中筛选出的6对核心引物具有较高的多态性;15个参试品种遗传相似系数变幅较大(0.35~0.91),其中8个选育品种及闽北闽南的4个参照种,相互间的遗传相似系数一般都达到均值以上(≥0.65),高者达0.80~0.91,证明其遗传差异小或较小;进行聚类分析,在相似系数为0.77处,可将参试材料分为8组,在相似系数为0.66处,可将参试材料分为6组;选用3对核心引物组合 W08、D07、L11建立15个参试品种的分子指纹图谱,特征条带大小210~630bp,可将每个品种逐一鉴别,效果较好。

Identification and preservation of tea cultivars plays a crucial role in the production and development of thetea industry in China.This paper presents the genetic variances and molecular fingerprints of 1 0 tea cultivarsselected and cultivated by our research team and 5 control cultivars in Fuj ian by using SSR molecular markers.Itaimed to further clarify the genetic differences of these cultivars providing a basis for identification of newlyimproved tea cultivars and protection of intellectual property rights.In the course,6 core primer pairs with highpolymorphism were selected from the 1 05 published pairs.The genetic similarity coefficient of the 1 5 tested cultivarsvaried greatly,ranging from 0.3 5 to 0.9 1. It generally reached above average (≥0.6 5)with high at 0.80-0.9 1 forthe 8 tea cultivars and 4 control cultivars from South and North Fuj ian among the tested varieties.Consequently,there was little or no genetic differences among the cultivars.Applying the cluster analysis,all tested materialscould be divided into 8 groups at the similarity coefficient of 0.7 7 ,and 6 groups in the similarity coefficient of 0.6 6.The molecular fingerprints of the tested cultivars were established using the primer pairs combination of W08,D07and L1 1. The characteristic stripes obtained were between 2 1 0-6 30 bp,which could conceivably suffice successfulidentifications of individual cultivars.
