目的:检测 Smad4和 VEGF蛋白在非小细胞肺癌(NSCLC)组织中的表达情况并分析其表达的相关性。方法采用免疫组织化学染色检测71例原发性 NSCLC及对应癌旁组织中 Smad4、VEGF蛋白的表达情况,分析Smad4蛋白表达与 NSCLC不同临床病理特征的相关性以及其与 VEGF蛋白表达的相关性。结果 Smad4和 VEGF蛋白在 NSCLC组织中的阳性表达率为57.7%(41/71)和71.8%(51/71),而对应癌旁组织中两者的表达率为77.5%(55/71)和49.3%(35/71),差异具有统计学意义(χ2=6.303,P=0.012;χ2=7.548,P=0.006)。Smad4蛋白表达与肿瘤分化(χ2=9.987,P=0.002)和TNM分期(χ2=5.943,P=0.015)有关。另外,NSCLC组织中 Smad4与 VEGF蛋白表达呈显著负相关(r=-0.638,P=0.009)。结论 NSCLC组织中 Smad4蛋白表达缺失,并与恶性临床病理特征密切相关,且 Smad4与 VEGF蛋白表达呈显著负相关,提示 Smad4缺失可能通过 VEGF促进肿瘤新生血管形成和肿瘤进展。

Objective To investigate the expression of Smad4 and VEGF and their correlation in human non-small cell lung cancer (NSCLC)tissues.Methods 71 samples of surgically resected NSCLC and matched normal tumor-adja-cent tissues were subj ected to immunohistochemistry for Smad4 and VEGF protein.Correlation between Smad4 protein expression and different clinicopathological features in NSCLC and its association with VEGF protein were analyzed. Results The positive expression rate of Smad4 and VEGF protein were 57.7% (41/71)and 71.8% (51/71),respec-tively,in NSCLC tissues,while their positive expression rates were 77.5% (55/71)and 49.3% (35/71),respectively, in matched normal-adjacent tissues.The difference was significantly (χ2=6.303,P=0.012;χ2=7.548,P=0.006). The expression of Smad4 protein was significantly correlated with tumor differentiation (χ2=9.987,P=0.002)and TNM tumor stage (χ2=5.943,P=0.015).Otherwise,Smad4 was inversely correlated with VEGF protein in NSCLC tis-sues (r=-0.638,P=0.009).Conclusion Loss of Smad4 protein expression is observed in NSCLC and its low-expression is associated with poor clinicopathological parameters.Furthermore,Smad4 is inversely correlated with VEGF protein in NSCLC, suggesting Smad4 loss may promote neoangiogenesis and tumour progression through VEGF in NSCLC.
