目的:研究炙甘草汤逆转快速起搏兔右心房(RAP)诱导心房颤动(AF)致右心耳心肌心肌纤维化的作用。方法新西兰大白兔动物模型随机分为假手术组(A组):植入电极不行快速起搏;起搏组(B组):行短期快速起搏兔右心房12 h致 AF模型;炙甘草汤水煎液阴性方组(C组):给予炙甘草汤阴性方水煎液灌胃2次/d,连续30 d后行短期快速起搏兔右心房12 h致 AF模型;炙甘草汤组(D组):给予炙甘草汤水煎液灌胃2次/d,连续30 d后行短期快速起搏兔右心房12 h致 AF模型;每组实验动物各8只。建立房颤兔模型,行兔右心耳 Masson染色和免疫组化方法检测基质金属蛋白酶-9(MMP-9)蛋白表达。检测右心耳组织场电位时程(fAPD),并观察炙甘草汤对右心耳 fAPD的作用。结果成功建立快速起搏右心房诱导房颤兔模型,Masson染色显示 B组和 C组心肌纤维化明显增加,D组心肌纤维化减弱。MMP-9结果显示,B组和C组 MMP-9呈强阳性,D组呈弱阳性。B组和 C组 fAPD明显缩短,差异有统计学意义(P<0.05)。给予炙甘草汤水煎液之后,可作用于房颤兔右心耳组织 fAPD导致 fAPD延长(P>0.05)。结论炙甘草汤可逆转房颤兔心肌纤维化,同时缩短 fAPD的时程。

목적:연구자감초탕역전쾌속기박토우심방(RAP)유도심방전동(AF)치우심이심기심기섬유화적작용。방법신서란대백토동물모형수궤분위가수술조(A조):식입전겁불행쾌속기박;기박조(B조):행단기쾌속기박토우심방12 h치 AF모형;자감초탕수전액음성방조(C조):급여자감초탕음성방수전액관위2차/d,련속30 d후행단기쾌속기박토우심방12 h치 AF모형;자감초탕조(D조):급여자감초탕수전액관위2차/d,련속30 d후행단기쾌속기박토우심방12 h치 AF모형;매조실험동물각8지。건립방전토모형,행토우심이 Masson염색화면역조화방법검측기질금속단백매-9(MMP-9)단백표체。검측우심이조직장전위시정(fAPD),병관찰자감초탕대우심이 fAPD적작용。결과성공건립쾌속기박우심방유도방전토모형,Masson염색현시 B조화 C조심기섬유화명현증가,D조심기섬유화감약。MMP-9결과현시,B조화C조 MMP-9정강양성,D조정약양성。B조화 C조 fAPD명현축단,차이유통계학의의(P<0.05)。급여자감초탕수전액지후,가작용우방전토우심이조직 fAPD도치 fAPD연장(P>0.05)。결론자감초탕가역전방전토심기섬유화,동시축단 fAPD적시정。
Objective To study the Zhigancao Decoction reducing right atrial myocardium of myocardial fibrosis based on rapid atrial pacing(RAP)in rabbits.Methods New Zealand white rabbits were randomly divided into sham operation group (A group)with implanted electrodes,no rapid pacing;pacing group(B group)with AF model induced by short-termrapid pacing rabbits right atrial 12 h;negative prescription of Zhigancao Decoction group(C group)with negative prescription of Zhigancao Decoction,Decoction orally 2times/d for 30 d,continuously then short-term rapid pa-cing of right atrial 12 h induced AF model;Zhigancao Decoction group(D group)with Zhigancao Decoction orally 2 times/d for 30 d,continuously then short-term rapid pacing of right atrial 12 h induced AF model.experimental animal of each group with 8 rats.Atrial fibrillation model established in rabbits.atrial myocardial fibrosis was tested by Masson staining and protein expression of matrix metalloproteinase-9 (MMP-9 )was detected with pathologic immunohisto-chemistry.right atrial appendage tissue field action potential duration(fAPD)was measured by microelectrode arrays (MEA).Results the rapid atrial pacing successfully inducted atrial fibrillation model in rabbit.Masson staining showed myocardial fibrosis significantly increased in B and C groups,myocardial fibrosis reduced in D group.protein expression of MMP-9 was strongly positive in B and C group,protein expression of MMP-9 was weakly positive in D group.fAPD significantly decreased in B group and C group,the difference was statistically significant(P<0.05).After administra-tion of Zhigancao Decoction,fAPD prolonged(P>0.05).Conclusion Zhigancao Decoction can reduce myocardial fibro-sis of atrial fibrillation in rabbits,and short the duration of fAPD.
