目的:研究岩大戟内酯B(jolkinolide B,JB)对内毒素血症模型小鼠的治疗作用。方法采用腹腔注射内毒素制备小鼠内毒素血症模型,观察不同脓毒JB(10、5和2.5 mg/kg)对内毒素血症模型小鼠的治疗效果。用ELISA法检测 LPS刺激后8 h小鼠血清中炎性细胞因子TNF-α、IL-1β和 IL-6的含量并检测腹腔灌洗液中白细胞数目。结果 JB以浓度依赖的方式降低内毒素血症模型小鼠死亡率,并显著降低血清中炎性细胞因子 TNF-α、IL-1β和 IL-6含量以及腹腔灌洗液中白细胞数目。结论 JB通过抑制内毒素血症小鼠炎性细胞因子表达以及炎性细胞浸润发挥其保护作用,可以成为内毒素血症治疗的候选药物。

목적:연구암대극내지B(jolkinolide B,JB)대내독소혈증모형소서적치료작용。방법채용복강주사내독소제비소서내독소혈증모형,관찰불동농독JB(10、5화2.5 mg/kg)대내독소혈증모형소서적치료효과。용ELISA법검측 LPS자격후8 h소서혈청중염성세포인자TNF-α、IL-1β화 IL-6적함량병검측복강관세액중백세포수목。결과 JB이농도의뢰적방식강저내독소혈증모형소서사망솔,병현저강저혈청중염성세포인자 TNF-α、IL-1β화 IL-6함량이급복강관세액중백세포수목。결론 JB통과억제내독소혈증소서염성세포인자표체이급염성세포침윤발휘기보호작용,가이성위내독소혈증치료적후선약물。
Objective To investigate the protective effects of jolkinolide B on mouse endotoxemia.Methods The mouse model of endotoxemia were prepared by intra-peritoneal inj ecting lipopolysaccharide (LPS)and the protective effects of JB at the concentrations of 10,5 and 2.5 mg/kg on mouse models of endotoxemia were investigated.Blood was collected to determine serum TNF-α,IL-1βand IL-6 levels by ELISA at 8 h after LPS treatment.The numbers of white blood cells were determined by hemocytometer count in the peritoneum wash.Results The mortality rate of mouse model of endotoxemia was decreased by JB pretreatment in a concentration-dependent manner.The numbers of white blood cells and the serum inflammatory cytokine levels such as TNF-α,IL-1βand IL-6 were decreased by JB in a dose-dependent manner.Conclusion JB exerts its protective effects on endotoxemia mouse by inhibiting the expression of inflammatory cytokines and inflammatory cells infiltration.The jolkinolide B will be the therapeutic drug candidate for endotoxemia..
