目的:观察苏黄泻浊丸对肾小球硬化大鼠转化生长因子-β1( TGF-β1)及其受体mRNA表达的影响,探讨其抗肾小球硬化的机制。方法:方法:采用单侧肾脏切除同时尾静脉注射阿霉素的方法建立肾小球硬化大鼠模型,SD大鼠随机分为空白对照组、模型组、参地补肾胶囊组、贝那普利组、苏黄泄浊丸组,按1 ml·100 g-1·d-1灌胃,实验结束后检测血肌酐( Scr)、尿素氮( BUN)、血浆白蛋白( Alb);免疫组化法检测TGF-β1表达,RT-PCR检测TGF-β1受体mRNA的表达水平。结果:苏黄泻浊丸能明显抑制TGF-β1及其受体mRNA的表达,治疗组与模型组比较,差异有统计学意义(P<0.01)。结论:苏黄泻浊丸能下调肾组织TGF-β1及其受体mRNA表达,改善肾脏病理形态,延缓肾小球硬化的进展。
目的:觀察囌黃瀉濁汍對腎小毬硬化大鼠轉化生長因子-β1( TGF-β1)及其受體mRNA錶達的影響,探討其抗腎小毬硬化的機製。方法:方法:採用單側腎髒切除同時尾靜脈註射阿黴素的方法建立腎小毬硬化大鼠模型,SD大鼠隨機分為空白對照組、模型組、參地補腎膠囊組、貝那普利組、囌黃洩濁汍組,按1 ml·100 g-1·d-1灌胃,實驗結束後檢測血肌酐( Scr)、尿素氮( BUN)、血漿白蛋白( Alb);免疫組化法檢測TGF-β1錶達,RT-PCR檢測TGF-β1受體mRNA的錶達水平。結果:囌黃瀉濁汍能明顯抑製TGF-β1及其受體mRNA的錶達,治療組與模型組比較,差異有統計學意義(P<0.01)。結論:囌黃瀉濁汍能下調腎組織TGF-β1及其受體mRNA錶達,改善腎髒病理形態,延緩腎小毬硬化的進展。
목적:관찰소황사탁환대신소구경화대서전화생장인자-β1( TGF-β1)급기수체mRNA표체적영향,탐토기항신소구경화적궤제。방법:방법:채용단측신장절제동시미정맥주사아매소적방법건립신소구경화대서모형,SD대서수궤분위공백대조조、모형조、삼지보신효낭조、패나보리조、소황설탁환조,안1 ml·100 g-1·d-1관위,실험결속후검측혈기항( Scr)、뇨소담( BUN)、혈장백단백( Alb);면역조화법검측TGF-β1표체,RT-PCR검측TGF-β1수체mRNA적표체수평。결과:소황사탁환능명현억제TGF-β1급기수체mRNA적표체,치료조여모형조비교,차이유통계학의의(P<0.01)。결론:소황사탁환능하조신조직TGF-β1급기수체mRNA표체,개선신장병리형태,연완신소구경화적진전。
Objective:To observe the effects of SuHuangXiezhuo pill on the expression of TGF-β1 and its receptor mRNA in rats with glomerulosclerosis. To investigate the mechanisms of anti glomerular sclerosis. Methods:Unilateral nephrectomy and in-travenous injection of adriamycin to establish the model of glomerular sclerosis rats, SD rats were randomly divided into blank control group, model group, shendi bushen capsule group, benazepril group,suhuang xiezhuo pill group, Press 1 ml·100 g-1 ·d-1 gavage, after the end of the experiment to detect serum creatinine ( Scr) , blood urea nitrogen ( BUN) , plasma albumin ( ALB) ,Immunohisto-chemical method was used to detect the expression of TGF-β1 , RT-PCR to detect the expression level of TGF-β1 receptor mRNA. Results:Suhuangxiezhuo pill could obviously inhibit TGF-β1 and its receptor mRNA expression. The treatment group Compared with model group. There are statistically significant differences (P<0. 01). Conclusion:Suhuangxiezhuo pill can inhibit the expression of TGF-β1 and its receptor mRNA, and improve the renal function, so that it may retard the development of glomerular sclerosis.
