目的:观察转录因子YY1(Yin-Yang 1)在垂体腺瘤中的表达情况,并进一步分析沉默YY1对大鼠垂体瘤GH3细胞侵袭性的影响。方法运用免疫组化技术观察YY1在不同类型垂体腺瘤组织中的表达情况。 Western blot实验检测siRNA沉默大鼠垂体瘤GH3细胞中的基因YY1后,YY1蛋白及基质金属蛋白酶MMP-2、MMP-9的表达水平。采用Transwell chamber细胞侵袭实验观察沉默YY1对GH3细胞侵袭的影响。结果 YY1在侵袭性垂体腺瘤中表达升高;沉默YY1后,GH3细胞侵袭能力下降35.7%(P<0.05), MMP-2、MMP-9蛋白表达量下降(P<0.05)。结论转录因子YY1在侵袭性垂体腺瘤中的表达升高,并通过上调MMP-2、MMP-9的表达来促进GH3细胞侵袭性。

Objective To investigate the expression of transcription factors YY1( Yin-Yang 1) in pituitary adenomas, and further analyse the effect of silencing YY1 on the invasiveness of rat pituitary tumor GH3 cells.Methods Using immunohistochemical technique, we observed the expression of YY1 in various type pituitary adenomas. The expression levels of YY1 protein and Matrix metalloproteinases MMP-2 and MMP-9 protein in rat pituitary tumor GH3 cells transfected with siRNA was measured by western blot .The invasion ability of GH3 cells after silencing YY1 by transwell chamber invasion assay.Results YY1 was increased in invasive pituitary adenomas.By silencing YY1, we found the invasion capability of GH3 cells were reduced by 35.7% (P<0.05), and the expression levels of MMP-2, MMP-9 were reduced(all P<0.05).Conclusion The expression of transcription factors YY1 is increased in invasive pituitary adenomas, which can promote the invasion of rat pituitary tumor GH3 cells by up-regulating the expression of MMP-2 and MMP-9.
