目的 研究透毒复方青蒿鳖甲汤对急性髓系白血病完全缓解期(AML-CR)患者骨髓CD34+细胞来源树突状细胞(DC)生物学效应的影响.方法 分离纯化骨髓CD34+细胞,体外不同浓度透毒中药含药血清与细胞因子联合诱导分化为DC,观察DC形态特征,流式细胞术检测DC表面分子CD80、CD83、CD86的表达,将DC分别与自体、异体外周血T细胞共同培养,四甲基偶氮唑蓝(MTT)比色法检测激活的T细胞不同效靶比对人类白血病细胞株(K562细胞)的杀伤效应.结果 青蒿鳖甲汤联合细胞因子能促进AML-CR患者骨髓CD34+分化为形态典型DC,不同剂量含药血清均能上调DC表面特征性分子及共刺激分子CD80、CD83、CD86比例,较单纯细胞因子组比较差异有统计学意义(P<0.01),且含药血清组较单纯细胞因子更能激发外周血T淋巴细胞对K562细胞的杀伤作用,两者间比较差异有统计学意义(P<0.01).结论 青蒿鳖甲汤能促进髓系微小残留白血病患者CD34+细胞向DC转化,提高DC的生物学效应.

Objective To study the effect of the serum with modified preparation of Toudu TCM Qinghao Biejia Soup to the induction of dendritic cell (DC) of complete remission acute myeloid leukemia (AML-CR) patients with source of CD34+ cells.Methods CD34+ cells from AML-CR patients were isolated,and cultivated combined with serums of different concentrations of Qinghao Biejia Soup.Transformation was observed under inverted microscope for the morphological characteristics of DC.DC surface molecules of CD83,CD80,CD86 expression were detected by flow cytometry.MTT method was used to detect the killing effect of leukemia cell line K562 in different conditions,induced transformation mature DC incubated with foreign or autolous T cells respectively.Results Qinghao Biejia Soup promoted AML-CR CD34+ cells differente to DC with typical morphology.Mature DC with high expression of specific markers (CD80,CD83,CD86) (P < 0.01).Conclusion Qinghao Biejia Soup can promote patients CD34+ cells to DC transformation.
