目的 探讨IL-21/STAT3通路在溃疡性结肠炎(ulcerative colitis,UC)小鼠肠道中的表达及其作用.方法 20只BALB/C小鼠随机分为正常组和DSS模型组各10只.小鼠UC模型用葡聚糖硫酸钠(dextran sodium sulphate,DSS)诱导,观察小鼠疾病活动指数(DAI)评分、病变结肠常规病理切片,采用ELISA法检测血清IL-6、IL-21含量,免疫组化检测结肠STAT3表达.结果 模型组小鼠DAI评分、血清IL-6和IL-21含量、结肠病理评分、黏膜STAT3表达水平均较正常组高,差异有显著性(P<0.05).结论 溃疡性结肠炎小鼠血清炎症介质及肠道STAT3表达水平明显升高,推测IL-21/STAT3通路可能参与UC小鼠肠道炎症损伤,阻断病变肠道IL-21/STAT3通路,减少STAT3蛋白表达或许是治疗UC的作用靶点.

목적 탐토IL-21/STAT3통로재궤양성결장염(ulcerative colitis,UC)소서장도중적표체급기작용.방법 20지BALB/C소서수궤분위정상조화DSS모형조각10지.소서UC모형용포취당류산납(dextran sodium sulphate,DSS)유도,관찰소서질병활동지수(DAI)평분、병변결장상규병리절편,채용ELISA법검측혈청IL-6、IL-21함량,면역조화검측결장STAT3표체.결과 모형조소서DAI평분、혈청IL-6화IL-21함량、결장병리평분、점막STAT3표체수평균교정상조고,차이유현저성(P<0.05).결론 궤양성결장염소서혈청염증개질급장도STAT3표체수평명현승고,추측IL-21/STAT3통로가능삼여UC소서장도염증손상,조단병변장도IL-21/STAT3통로,감소STAT3단백표체혹허시치료UC적작용파점.
Objective To approach the role and expression of IL-21/STAT3 pathway in the pathogenesis of murine with ulcerative colitis(UC).Methods 20 BALB/C mice were randomly divided into normal group and DSS model group,10 mice in each group.UC mice were induced by drinking dextran sodium sulfate solution.The mouse disease activity index score(DAI score)was calculated.Mice were killed at the eighth day,the colon tissues were conducted with pathological section and score,serum content of IL-6 and IL-21 were assayed by Elisa kits,the expression of STAT3 protein in colonic mucosal was detected by immunohistochemical method.Results The score of DAI and histological damage score,serum IL-6 and IL-21 content and the expression level of STAT3 in colonic mucosal were higher in model group than those in normal group(P < 0.05),the difference had statistical significance(P < 0.05).Conclusion The serum contents of IL-6 and IL-21 and the expression level of STAT3 intestinal tract were increased in mouse with UC,we presume that IL-21/STAT3 pathway may participate in the process of UC,blocking IL-21/STAT3 pathway in illness intestinal tract and reducing STAT3 expression may be the drug treatment target of UC.
