目的 分析基质细胞衍生因子-1(SDF-1)及其受体CXCR4、凋亡抑制蛋白基因livin在肺癌组织中的表达与肺癌临床特征的关系,并探讨各基因表达的相关性.方法 用逆转录多聚酶链式反应检测52例肺癌组织、16例癌旁正常肺组织及3例肺部良性病变组织中SDF-1、CXCR4及livin mRNA的表达.结果 肺癌组织中SDF-1的表达率为90.38% (47/52),显著高于癌旁正常肺组织(37.50%,6/16)及肺部良性病变组织(33.33%,1/3)(P=0.000;P=0.040).CXCR4在肺癌组织中的表达率为88.46% (46/52),显著高于癌旁正常肺组织(25.00%,4/16)及肺部良性病变组织(0.00%,0/3)(P=0.000;P=0.003).livin在肺癌中的表达率为84.62%(44/52),显著高于癌旁正常肺组织(18.75%,3/16)及肺部良性病变组织(0.00%,0/3)(P=0.000;P=0.001),其中livinα表达阳性率为73.08%(38/52),livinβ为69.23%(36/52).SDF-1、CXCR4及livinα、livinβ mRNA在肺癌组织中的表达与肺癌病理类型、分化程度、肿瘤部位、肿瘤大小、是否发生淋巴结转移、TNM分期及有无化疗均无明显相关关系(P值均>0.05).肺癌组织中CXCR4仅与livinα呈正相关(r=0.287,P=0.039),SDF-1与livinα及livinβ均无明显相关关系(P=0.311; P=0.129),SDF-1与CXCR4亦无相关关系(P=0.460).结论 SDF-1、CXCR4及livin在肺癌组织中高表达,参与肺癌的发生发展;CXCR4与livinα密切相关,但与肺癌是否发生淋巴结转移无明显相关关系.

Objective To investigate the expressions and clinical significances of SDF-1,CXCR4,and livin in human lung cancer,and to explore the correlations between these genes.Methods Reverse transcription polymerase chain reaction was performed to detecte the expressions of SDF-1,CXCR4,and livin mRNA in 52 lung cancer tissues,16 normal lung specimens taken from non cancerous regions adjacent to lung cancer tissues,and three benign disease lung tissues.Results The positive rate of SDF-1 in lung cancer tissues (90.38%,47/52) was much higher than that in para-cancerous (37.50%,6/16) and benign disease lung tissues (33.33%,1/3) (P =0.000,P 0.040).The positive rate of CXCR4 in lung cancer tissues (88.46%,46/52) was much higher than that in para-cancerous (25.00%,4/16) and benign disease lung tissues (0.00 %,0/3) (P =0.000,P=0.003).In lung cancer tissues,the positive rates of livinα and livinβ were 73.08%(38/52) and 69.23%(36/52) respectively.Besides,the positive rate of livin (84.62%,44/52) was also higher than that in para-cancerous (18.75%,3/16) and benign disease lung tissues (0.00%,0/3) (P=0.000,P =0.001).There were no correlations between the expressions of SDF-1,CXCR4,livin and clinical characteristics (pathological types,differentiation grade,location,size,lymphnode metastasis,TNM stage,chemotherapy before operation)in lung cancer tissues (all P >0.05).The expression level of CXCR4 was positively associated with that of livinα in lung cancer tissues (r =0.287,P=0.039),but there were no correlations between SDF-1 and living,livinβ,CXCR4 (P =0.311,P =0.129,P =0.460).Conclusions SDF-1,CXCR4,and livin are highly expressed in lung caner tissues,participate in the occurrence and development of lung cancer.CXCR4 is intimately associated with livinα,however,there is no significant correlation between CXCR4 and lymphnode metastasis in lung cancer tissues.
