目的 研究大鼠在高原低氧环境下肺动脉高压形成过程中水通道蛋白-1(aquaporin-1,AQP-1)表达的变化和意义.方法 将60只200~280 g的雄性Wistar大鼠分为3组,对照组1组和实验组3组;对照组为平原对照组;实验组分别为高原低氧环境10 d、20 d、30 d组.高原低氧组大鼠置于模拟海拔5 000 m低压低氧舱,每天22 h,对照组大鼠在平原环境饲养;应用西门子6002有创血压监测系统,测定平均肺动脉压(MPAP),而后取出肺组织标本做免疫组织化学染色.结果 ①MPAP:高原低氧10d、20 d、30 d组与对照组比较P值均<0.05;②AQP-1:高原低氧10 d、20 d、30 d组与对照组比较,P <0.05;③相关性分析:大鼠MPAP、AQP-1呈正相关.结论 AQP-1在高原低氧环境下表达增高,与肺动脉压的严重程度相关.

Objective To study the expression and singnificance of AQP-1 in lung tissue of rats with hypoxic pulmonary hypertension.Methods 60 Wistar male rats which weight rang from 200 g to 280 g were divided into four groups:control group,altitude hypoxia environment 10 d,20 d,30 d group,measured the pulmonary artery pressure of each group and AQP-1 immunohistochemical staining in lung tissue,analysis the expression AQP-1 and the relationship with pulmonary artery pressur.Results The expression intensity of AQP-I and pulmonary artery pressure gradually increased with altitude hypoxia time extension,the expression of AQP-1 and the pulmonary artery pressure slowly increase with time extent.Conclusions AQP-1 and pulmonary artery pressure was positively correlated at the process of altitude hypoxia environment pulmonary hypertension,AQP-1 may play an important role in the development of pulmonary hypertension of hypoxia environment,by regulating the expression of AQP-1 may prevent pulmonary hypertension.
